The downyfeather of goose has an important economic value,throughput and quality of downyfeather depends on the growth of feather follicles.The occurrence and development of goose primary and secondary feather follicles were in the embryonic stage.Especially in late embryos,along with the nutritional requirements increase of embryos,The development of feather follicle is also affected.Methionine is the first restrictive amino acid in poultry.Therefore this experiment studied the developmental of primary and secondary follicles from late embryos to born 28 d of Jilin white goose,and the influence of in OVO feeding Methionine(Met)on feather follicle growth performance and relative expression quantity of gene IGF-1 and gene EGF in Jilin white goose.1.Influence of Jilin white goose feather follicle development and in OVO feeding Met on feather follicle developmentThere were 300 Jilin white goose fertile eggs were used in this experiment.All eggs were randomly into two treatments(CON and TRT)and each treatment had 6 repeats to hatch.At the 24 d of incubation,all TRT eggs were injection 1.5 ml Met nutrient solution including Met 5 mg/ml and NaCl 7.5 mg/ml,CON treatment no injection.Feeding experiment was 28 days from hatched.The skin samples form backside,chest and abdomen were taken on the day E23? E27? 0d? 1d? 4d? 7d? 14d? 21d? 28 d,and then paraffin sections were made to measure diameter and density of featner follicle.Here is the results: E23 to 28 d,there was significant(P < 0.05)lower of primary follicle density on backside,chest and abdomen,at the same time,there was significant(P < 0.05)higher on secondary follicle density on the same part.Primary and secondary follicle density on chest were higher than their on backside and abdomen.The difference of primary feather follicle diameter on the three part were as follow: E23~E27,significantly(P < 0.05)increase;E27~14d,no significant(P > 0.05);14d-28 d significant(P < 0.05)increase.The difference of secondary feather follicle diameter on the three part were as follow: E27~28d,significantly(P < 0.05)increase.Primary and secondary follicle diameter on backside were higher than their on chest and abdomen.All the indices including primary feather follicle density,secondary feather follicle density,primary feather follicle diameter and secondary feather follicle diameter on backside,chest and abdomen from goose were significant(P < 0.05)higher from TRT to CON.From these results we can infer that primary feather follicle maybe were well development at the late embryos stage,the most important developing time for Jilin white goose secondary feather follicle were in the embryonic stage,chest was the main part of downyfeather produce.In OVO feeding Methionine can promote the growth of primary and secondary feather follicle in Jilin white goose.2.IGF-I and EGF gene regulate on Jilin white goose feather follicle and effect of in OVO feeding methionine to the regulation.This experiment to detect the relative expression of mRNA two gene IGF-I and EGF in Jilin white goose skin by using real-time fluorescence quantitative PCR method,which shows that IGF-1 and EGF two gene regulation of hair follicle development.The results showed that the expression of IGF-I gene was expressed during the embryonic and postnatal development of feather follicles,from E23-28 d the expression of IGF-I on skin gradually increased,E23-E27 no significant(P > 0.05)increase,E27-7d increased significantly(P < 0.05),7d-14 d no increased significant(P > 0.05),14d-28 d increased significantly(P < 0.05).The EGF gene was expressed in the hair follicle during embryonic and postnatal development,E23-E27 had relatively stable expression around 1.00.Form E27 to 28 d,there were a increasing expression,E27-14 d significant(P < 0.05)increase and 14d-28 d increased no significant.E27,0d,7d,and 28 d TRT had significant(P < 0.05)higher expression on gene IGF-1 and EGF.Form these result I can infer that IGF-I and EGF gene can regulate the growth of Jilin white goose feather follicle,this two gene can promote the growth of secondary feather follicle.In late embryos,the most growth of secondary feather follicle stage,in OVO feeding Met,promote feather follicle development by increasing the expression of IGF-1 and EGF two gene.
