Vaginal microbiota plays a critical role in guarding against infections in the reproductive tract by pathogenic bacteria.At present,the most common practiced treatment method against the reproductive tracts infections is the use of antibiotic therapy.However,antibiotic therapy has prominent side effects such as microbial resistance and antibiotic residues in milk.These challenges poses an urgent attention to develop an alternative method to treat reproductive tracts infections without side effects.The aim of this study was isolated lactobacillus from postpartum healthy dairy cow vaginas to provide strains for developing microecological preparations to prevent and treat endormetritis of dairy cows.The vaginal Lactobacillus were isolated by Lactobacillus agar culture and selected by antimicrobial activity and identified by 16 S rDNA sequencing analysis.The ability of Lactobacillus to produce acid and H2O2 were tested,the susceptibility to antibiotics of Lactobacillus were evaluated by Disk diffusion test and analyze anti-bacteria substance.The best probiotic candidates were screened out by the above probiotic function,and the clinical effect of Lactobacillus on endometritis was studied by establishing the pathology model of rabbits.30 Lactobacillus isolates were obtain in the research,and 7 isolates were selected by anti-bacteria activity which have the ability of inhibit growth of pathogenic bacteria.According to the 16 S rDNA sequence identification,the 7 Lactobacillus isolates were 2 Lactobacillus salivarius?2 Lactobacillus mucosae? 1 Lactobacillus reuteri?1 Lactobacillus gasseri and 1 Lactobacillus johnsonii.Seven Lactobacillus isolates had good producing acid property and were susceptible to three antibiotics above at last,and most Lactobacillus can produce hydrogen peroxide.The anti-bacteria substance of Lactobacillus johnsonii LWT11 were organic acid was major antibacterial substance for lactobacillus to inhibit the growth of Staphylococcus aureus and E.coli and Salmonella,Rabbits are cured of uterus in pathologic model,which are injected into vagina with culture fluid of combination of Lactobacillus salivarius LWT1 and Weissella cibaria for 4 time.Result shows that 80% of effective rate,60% of cure rate.In conclusion,combination of Lactobacillus salivarius LWT1 and Weissella cibaria preparation of microecological agents can be used to treat endometritis caused by intrauterine perfusion of E.coli,which can be used for preventing and treating of postpartum endometritis of dairy cows.
