Cyprinus carpi,Carassius auratus,Ctenopharyngodon idellus and Onychosotoma gerlachi,all of which belong to Cyprinidae and Cypriniformes,were economically important fish in the Duliu River,Guizhou,China.The living environment and population continuation of this four species have been damaged to varying degrees due to enhancement of human fishing activities,the establishment of coastal factories and the construction of water conservancy projects.At present,Studies on the germplasm resources of this four species in the Duliu River were deficiency.Therefore,the genetic structure of mtDNA D-loop and Cyt b gene of this four species used as the research object were analyzed by DNA sequencing and PCR technology and their present status of germplasm resource was studied and evaluated,in order to provide the theoretical basis for the utilization and protection of the Cyprinidae economic fish in Duliu River.The main results were as follows:1 ? In this study,from Cyprinus carpi that collected from Duliu River,we obtained the length of mtDNA D-loop and Cyt b were 899 ~ 901 bp and 1133 bp.Results showed that in the two sequences,A+T was higher than C+G,and the content of G was lowest.In the study,we detected the extended termination associated sequence(ETAS)?central conserved domain(CD)and the conserved sequence block(CSB).In the sequences of D-loop,a total of 24 polymorphic loci were found.40 individuals belonged to 12 haplotypes.Types of Mutations: Conversion,Substitution,Deletion and Insertions.In Cyt b sequences,a total of 28 polymorphic loci were found.There were 22 synonymous and 6 non synonymous mutation sites,and mainly distributed in the first codon.15 haplotypes were defined with only base transformation and transversion.The genetic diversity parameters of mtDNA D-loop and Cyt b gene showed that thepopulations with high genetic diversity.2?The mtDNA D-loop and Cyt b sequences of Cyprinus carpio in Duliu River were 787 bp and 1140 bp respectively.The content of A/T was higher,and G was the lowest.In this study,we identified mtDNA D-loop's termination region,central conserved region and conserved sequence region.A total of 24 polymorphic loci were found in the D-loop sequence.43 individuals belonged to 17 haplotypes.Mutation types were Transition or Transversion,no Deletion and Insertion detected out.In the Cyt b gene,a total of 25 polymorphic loci were found out.Including 23 synonymous mutation,2 non synonymous mutations,and mainly distributed in the first codon.10 haplotypes were defined,all were Transition or Transversion.The mtDNA D-loop and Cyt b gene parameters of genetic diversity showed that the genetic diversity of Cyprinus carpio was higher.3 ? This research has obtained the gene length of Ctenopharyngodon idellus,mtDNA D-loop 901~905bp and Cyt b 1140 bp.The A/T content was higher than C/G and G base content was the lowest in both sequences.In this study,we also distinguished ETAS?CD and CSB of mtDNA D-loop.A total of 11 polymorphic loci were found out.38 grass carp belonging to 8 haplotypes.Variant types include Conversion,Transversion,Deletion and Insertion.In Cyt b of Ctenopharyngodon idellus population,a total of 10 polymorphic loci were detected.8 synonymous mutation,2 non synonymous mutations,and mainly distributed in the first codon.8 haplotypes were defined in total.The type of mutation was Conversion and Transversion.Parameters results indicated that the population genetic diversity was low.4?In the study,we has obtained the mtDNA D-loop(686~687bp)and Cyt b(1140bp)gene sequences of Onychosotoma gerlachi.The content of both two genes A+T was higher than C+G,and the content of G was the lowest.In this study,we distinguished ETAS?CD and CSB,but did not detected CSB-2 and CSB-3.A total of11 polymorphic loci were found in the D-loop sequences,belonging to 15 haplotypes.Variant types include Conversion,Substitution,Deletion and Insertion.In the Cyt b gene,a total of 3 polymorphic loci including 2 synonymousmutation and 1 non synonymous mutations were detected out.Mutation sites were all located in the third codon.A total of 4 haplotypes were defined.Variant types include Transition and Transversion.The genetic diversity parameters results given an indication that the population was lack of germplasm resources.
