Changshun blue-shelled chicken was a local breed of Guizhou Province.It was necessary for us to purify Changshun green-shelled chicken in order to protect and use the poultry resource preferably.In this study,we selected 1690 Changshun blue-shelled chickens randomly.The oocyan gene(Oocyan,O)was as a candidate gene,three specific primers were designed,and we screened out dominant homozygous genotype of Changshun green-shelled chicken by multiplex PCR technology.In order to study the difference expression of HO-1gene in different tissues of 70-weeks-old Changshun blue-shelled chickens,we collected nine(3 of each genotype)Changshun blue-shelled chickens of 70-weeks-old as samples.The brain,liver,kidney,ovary,fallopian tube and uterus were collected,and total RNA of every tissue was extracted and reversed transcription to cDNA.The expression of HO-1 mRNA in different samples were detected by quantitative real-time PCR.We compared the expression in the same tissues of different genotype and in the different tissues of the same genotype.And,we detected the concentration of biliverdin in three different genotypes of eggs.The results showed that:1.The dominant homozygous genotype was 423 in the experimental group,the recessive homozygous genotype was 153,and the heterozygous genotype was 1114.We found that the gene was belong to moderate polymorphism by the genetic analysis,and it wasn't reached to Hardy-Weinberg equilibrium state in this population,this may be related to the certain selection of the group.The frequency of heterozygous genotype in the group was 67.21%,and the offspring of heterozygote would appear the segregation of character.Therefore,it was necessary to improve the frequency of dominant homozygous genotype and the frequency of dominant gene so as to improve the rate of oocyan eggs and increase economic benefits.2.The differences were significant among different tissues of the same genotype of HO-1 gene(P<0.05).And the differences were also significant among different genotypes of HO-1 gene in the same tissue(P<0.05).The expression of HO-1 gene was highest in the liver,and there were significant difference in different genotypes(P<0.01).The expressionlevel in the uterus of white-shelled chicken was significantly higher than the other two genotypes(P < 0.01),there was no significant difference between homozygous and heterozygous blue-shelled chickens(P>0.05).3.The differences were significant among different tissues of the same genotype of SLCO1C1 gene(P < 0.05).And the differences were also significant among different genotypes of SLCO1C1 gene in the same tissue(P<0.05).The expression of SLCO1C1 gene was highest in the oviduct,and there were significant difference in different genotypes(P<0.01).The expression level in the ovary of white-shelled chicken was significantly higher than other two genotypes(P<0.01),there was no significant difference between homozygous and heterozygous blue-shelled chickens(P>0.05).4.The concentration of biliverdin of eggs was highest significant difference among three different genotypes(P<0.01).The concentration of biliverdin of the dominant homozygous genotype individuals was significantly higher than other two genotypes.
