Dendrobium officinale Kimura et Migo,belonging to Dendribium Sw.(Orchidaceae),is a perennial herbaceous plant.It is regarded the best medicinal materials of the world because of its role that it can nourish your Yinjin,protect your liver and bravery,benefit to your spleen and stomach.But the D.officinale has a wide distribution in China.The content and types of its main chemical components are different because of its special habitat.Through fingerprint technology and the test of the polysaccharide and alcohol soluble extract content,we can explore the difference of chemical constituent types and content of D.officinale of different origin,different germplasm,different morphology,different body part and different harvest time.The main results were as follows:1.Established the HPLC chromatographic analysis condition of D.officinale fingerprint.Taking the method of extracting the constituents that dissolved in 95%alcohol to treat the sample of D.officinale then concentrate it to 5 mL,filter it with the 0.22?m microporous membrane.Take the acetonitrile(A)and 3%phosphoric acid solution(B)as mobile phase to carry out the gradient elution.And the gradient elution program were as follows:the gradient time 0?5?25?60?105?139?165 min,and the corresponding concentration gradient of acetonitrile(A)is 5%?5%?10%?15.5%?24%?33%?39%;The detection wavelength is 210 nm and flow rate is 1.0 m L/min;and the chromatographic column:UltimateXB-C18(250 mm×4.6 mm,5?m).The elution conditions is suitable for almost all the samples that to establish the fingerprint of D.officinale.2.Established the alcohol soluble extract fingerprint of D.officinale grown in different area and analyzed its effective component content.(1)Established the fingerprint of D.officinale grown in Guangdong,Zhengjiang,Yunnan and so on of 10 provinces.There are27 common peaks which can be used as the characteristic peak to identify the D.officinale.The similarities between the 10 provinces are 0.229~0.861,the difference is larger.Through clustering analysis,the D.officinale origin can be divided into 2 categories.The polysaccharide and the alcohol soluble extract content measured have shown that the region where can produce better quality D.officinale are Zhejiang,Anhui,Yunnan,Jiangxi and Hunan.(2)There are 67 common peaks in 5 regions of Yunnan,and the similarity is 0.712~0.880.The region of D.officinale can be divided into two categories.The city of Baoshan rank first according to the polysaccharide and the alcohol soluble extract content standard;There are 69 common peaks in 4 regions of Zhejiang province,and the similarity is 0.698~0.775.The region of D.officinale can be divided into two categories.The polysaccharide and the alcohol soluble extract content of D.officinale grown in Leqing and Hangzhou have reached the standard authorized by the Chinese Pharmacopoeia.(3)There is a specific negative correlation between the content of polysaccharide and alcohol soluble extract.If the polysaccharide content is high,the extract content is relatively low.3.Established the alcohol soluble extract fingerprint of D.officinale of different germplasm and different body part of the same germplasm,then analyzed its main component.(1)Each code of D.officinale has their specific chromatographic peak at certain retention time.T6 and T701 at the time t _R=136.5 min,138 min,142 min,144 min have chromatographic peaks of big peak areas respectively,but T636 vice versa;As for T6,it has a specific peak that can differ from other code of D.officinale at the time t_R=160min.T636 at t _R=18 min,89 min has two obvious characteristic peaks;the polysaccharide and extract content of the three code D.officinale all reached the standard of Chinese Pharmacopoeia,and the polysaccharide content of T636 was the lowest(28.88%)while its extract content was the highest(8.29%).(2)The number of the chromatographic peaks of the leaf is similar to that of the stem,but the content is higher than the stem.The chemical constituents of the leaf differ widely from that of the stem between 45 min<t_R<70 min.(3)The fingerprint picture of the red stem and the green stem differes widely.We suggest to grow more red stem D.officinale after analysised the content of polysaccharide and alcohol soluble extract.4.Studied the alcohol soluble extract fingerprint of D.officinale of different harvest time.And find that the total peak area,peak number,alcohol soluble extract content of D.officinale presents such a change rule as from Nov.began to decline and in steady state fluctuations between Mar.and May.of the next year,then at Jul.take into an upward trend.But polysaccharide content performances basically the opposite trend.5.Studied the alcohol soluble extract fingerprint of D.officinale,D.nobile,D.loddigesii,D.devonianum and their effective component content as well.The difference among the 4 kinds of Dendrobii is great.They have 22 common peaks.The similarity among them is 0.061~0.217.D.loddigesii has its own specific peaks at the time t _R=17.5min,92 min,105 min,116 min,162 min;the distinct peak's retention time of D.nobile is t_R=13 min while the D.officinale t_R=160 min.And according to the retention time of the characteristic peaks we can distinguish the four kinds of Dendrobii.
