| Using culturable method,the rhizosphere multifunctional probiotics-Streptomyce-s S506 and 12 years of continuous cropping tomato soil as test material,taking samples at the tomato seedling stage,flowering and fruiting period,studied at the endophytic bacteria population structure and dynamic changes of the density of endophytic bacteria isolated in roots and stems,and founding the functional resources which could antagonize pathogens,produce IAA and siderophore、dissolve organic phosphorus and inorganic phosphorus and have the nitrogen fixation.biological assay has been used for the predominant strains.1.Screened a function complementary media combinations-nutritious medium NR and oligotrophic medium R2A,the two medium combination separable endogenous bacterial counts increased by 20.97%than the classic NA medium bacterial species by 160%.2.166 endophytic bacteria had been isolated from the different growth stages of tomato roots organization.Using 16S rDNA sequence analysis method to clear their phylogeny respectively belong to 30 kinds,Including Bacillus 14 species,Microbacterium 3 species,the Pantoea 2 species,Staphylococcus 3 species and eight kinds of other strains.Wherein,the bacterial strain numbered YT82 system developed with the nearest Bacillus foraminis similarity is only 95%,is likely to be an new resource of endogenous bacterial.3.S506’s intervention of tomato roots led to a significant change of he dynamic changes of the total population structure and advantage bacterium group of culturable endogenous bacteria at different growth stages of tomato.(1)Endogenous total bacteria was significantly improved in the flowering stage,the total difference of root and stem tissue endogenous bacteria were increased by 153.70%and 118.78%,the difference was highly significant level(p<0.01).Other period did not change significantly.(2)Endogenous bacterial population diversity increased significantly in the flowering stage.After S506 processing endophytic bacteria in the root and stem tissue,respectively,the population of bacterial increased of 133.33%and 160%.Other periods did not differ significantly.(3)The population of functional bacterial significantly increased.the population of functional bacteria in root which produced IAA and siderophore、dissolved organic phosphorus,have the nitrogen fixation and have the ability to antagonize pathogens increased of 156.3%,70%,155.6%,140%,100%and 63.2%.This result showed highly significant in promoting root growth,improving plant biomass and significantly improving disease prevention feature with the S506’s strain involved in the rhizosphere.(4)Roots advantage bacterial flora dominated by a single Bacillus amyloliquefaciens to diversify shift.In the flowering stage,in control roots,Bacillus amyloliquefaciens Account for 61%of the total separation,t S506 processing distribution tends to diversification,Bacillus subtilis Account for 29%,Bacillus amyloliquefaciens,Bacillus licheniformis and Bacillus megaterium Account for 16%each.The stem tissue dominant flora turned from Bacillus megaterium single dominant to Bacillus amyloliquefaciens and Bacillus subtilis.(5)S506 can inhibit Providencia vermicola,Staphylococcus saprophyticus and Klebsiella pneumoniae as human and animal pathogens and Ice nucleation bacterna-Pantoea agglomerans and Pantoea ananatis in tomato tissue colonization.4.Took winter wheat seed for the test material,screening for 5 functional bacteria which significantly promote germination and seedling growth,this 5 bacteria all isolated from the the S506 treatment plant.these bacteria can germinate wheat seeds early 3h,make plant and root dry weight increase by more than 52%and 70%respectively,leaves and root length has improved significantly.The 5 functional bacteria ith greater potential. |
