| This paper studied the effects of different dose of tannin in diet and high tannin diet supplemented with PEG on the quantity and diversity of ruminal bacteria in sheep and cashmere goats. Four sheep and four cashmere goats, age of 1.5-2-years old, weight of 45kg, fitted with permanent ruminal cannula, were used as experimental animals. The study was conducted in consecutive four periods, including control periods (basal diet, I), experimental period II (basal diet+2% tannin), experimental period III (basal diet+6% tannin) and experimental period IV (basal diet+6% tannin+12%PEG). Each period lasted for 30 days consisted of a 12-day pre-test period and 18-day test period. Ruminal content was collected before feeding and 6h after feeding, real-time PCR was used to quantify total bacteria, fungi, R.albus, R.flavefaciens, F.succinogenes, total methanogens, streptococcus bovis, prevotella ruminicola and butyrivibrio fibrisolvens adhered to solid phase, liquid phase and full chyme. In addition, DNA from rumen full chyme was extracted and 16s rRNA sequencing technique was used to analysis diversity of ruminal bacteria.The results showed that:1) As compared with group I, the population of general bacteria, anaerobic fungi, R.albus, R.flavefaciens, F.succinogenes, prevotella ruminicola, butyrivibrio fibrisolvens and methanogens adhered to rumen full chyme of group II and III were significantly decreased at Oh (P<0.05). However, at 6h the quantity of general bacteria, R.flavefaciens, F.succinogenes, prevotella ruminicola, butyrivibrio fibrisolvens and methanogens adhered to rumen solid phase of group II and III were significantly lower than those of group I (P<0.05).2) After supplementation of PEG (group IV), the population of general bacteria, anaerobic fungi, R.albus, R.flavefaciens, prevotella ruminicola and streptococcus bovis adhered to rumen full chyme at Oh were significantly higher than those of group II (P< 0.05). At 6h, the quantity of general bacteria, anaerobic fungi and R.flavefaciens adhered to rumen full chyme of group â…£, as well as Esuccinogene, prevotella ruminicola and methanogenes adhered to rumen solid phase of group IV were significantly higher than those of group II and III (P<0.05).3) The quantity of anaerobic fungi, R.flavefaciens, streptococcus bovis and butyrivibrio fibrisolvens adhered to rumen full chyme of sheep were significantly higher than those of goats at Oh (P<0.05). However, at 6h the quantity of R.flavefaciens and methanogenes adhered to rumen solid phase of goats were significantly higher than those of sheep (P<0.05).4) The differences among treatments in ruminal bacteria diversity and abundance were observed. The diversity of ruminal bacteria was highest in group â…¡ and lowest in group â… in sheep and goat.At-the same time, we observed that diversity and abundance of ruminal bacteria in sheep were higher than those of goats.5) Taxonomy results indicated that bacteroidetes and firmicute were predominant ruminal bacteria in four treatment groups of sheep and goat. As compared with group â… , relative abundance of bacteroidetes in group â…¡ and firmicute in group â…¢ decreased, whereas relative abundance of firmicute in group â…¡ and proteobacteria in group â…¢ increased. At genus level, relative abundance of prevotella was highest in four treatments, and relative abundance of prevotella and fibrobacter of sheep and goats in group â…¡ were lower than those of group â… , and the proportion of Rc9-gut-group in group â…¡ were higher than those of group â… . |
