In this experiment,toon was selected by the Fujian Provincial Forestry Academy as materials to study tissue culture,changes of flavonoids and polyphenol content in different growing leaves,antioxidant capacity of extracts in vitro,the impact on Drosophila life and identification of volatile chemical constituents.The results were as follows:(1)study on toon tissue culture demonstrated that the optimal proliferation medium was MS+BA1.0mg/L+ NAAO.1mg/L+GAO.1mg/L;the best rooting medium was MS+NAA0.2mg/L+IBA0.2mg/L;Humus soil can be used as transplanting substrate.(2)in different month,content of flavonoids,polyphenol and the DPPH free radical scavenging ability of Toona sinensis leaves(TSL)were measured.The results indicated flavonoid content increased at first and then decreased,there are significant differences between growth stages;polyphenol content upward over time,there are significant differences in old leaves and young leaves.Toona sinensis extracts has effect on DPPH radical scavenging capacity stronger than a single vitamins E.(3)Drosophila life was extended by certain concentration TSL extract,the best extract concentration for female flies was 0.1%,the best concentration of the extract for male flies was 1%;with increasing of concentration of TSL extracts,soluble protein content increased,SOD activity was significantly increased and lipid peroxidation product MDA content decreased in Drosophila,and showed good dose-response relationship.(4)life span effects of TSL extracts on Alzheimer Drosophila was not significant.(5)There are 36 kinds of volatile components in toon leaves,including2,4-dimethyl-Thiophene(18.52%),1-Calamenene(6.9%),.beta.-Cadinene(2.29%),etc;37 kinds of volatile components were found in flowers,mainly includs Caryophyllene(8.81%),(laR)-1aβ,2,3,3a,4,5,6,7bβ-Octahydro-1,1,3aβ,7-tetramethyl-1H-cyclopropa[a]naphthalene(7.88%),Eremophilene(7.61%),(-)-beta.-Cadinene(5.90%),.alpha.-Cubebene(4.15%),Z,Z,Z-1,5,9,9-tetramethyl-1,4,7,-Cyclou ndecatriene(3.94%),Isocaryophillene(3.45%),I-Calamenene(2.96%),etc;and 26 kinds of volatile components in seeds were identified,high content components including(-)-a-Selinene(15.37%),10s,11s-Himachala-3(12),4-diene(15.000%),alpha.-Cubebene(5.18%),(-)-beta.-Elemen(4.13%),d-Cadinene(3.87%),1,2.4a,5,6,8a-hexahydro-4,7-dimethyl-1-(1-methylethyl)-Naphthalene(3.72%),(-)-Germacrene D(2.89%),etc.There were four compounds were coexisting in the three different organs.The study proved that toon has good antioxidant capacity and provide scientific data for the further study and applying. |
