With the progress and development of science and technology,high-throughput sequencing technology in terms of transcriptome studies have been very widely used.Known as the "kingdom of plants" of Yunnan,its unique natural growing conditions,has become a veritable "fungi kingdom".In this study,we complete Yunnan wild edible and medicinal fungi's transcriptomes sequencing of Boletaceae,Leccinum,Leccinum extremiorientale and Boletaceae,Boletus,Boletus bicolor with Illumina/HiseqTM 2000 sequencing platform,and then completed the denovo assembly work for the transcriptomes of the fungi,and they were further completed the comparison and analysis of transcriptomes,gene functional annotation for transcriptomes.In this study,Leccinum extremiorientale and Boletus bicolor were identified out of 2,730 SSRs and 1,652 SSRs,respectively.Then we take advantage of CandiSSR to make further analysis.We make the Boletus bicolor transcriptome-SSRs to be reference sequence,and then blast the Leccinum extremiorientale transcriptome-SSRs to the reference sequence.We finally identify 89 SSRs with polymorphic locus.At the same time,we extract 100bp with upstream and downstream for designing primers,respectively.By comparing with these two fungi' transcriptomes,their orthologous gene suffered selective pressure during evolution was studied,we find that there are 52 orthologous genes may have a positive selection and 6 orthologous genes may have a strong positive selection of them.Finally,we complete the annotation and comparative study of carbohydrate enzyme related genes with fungi.It will contribute to functional genomic studies and Laid base of the foundation for further study of metabolic,pharmacological effects and characteristics of these datasets and results.
