Diarrhea is a worldwide public health problem caused by a pathogen and multi factor.Its incidence are very high and have great harm to animal health.Diarrhea is one of the major causes of malnutrition and death.Diarrhea is the leading cause of death in many developing countries,and the international WHO has listed the control of diarrhoeal diseases as a global strategy.At present,the main drugs for the treatment of diarrhea are antibiotics,but the side effects caused by the use of antibiotics have become increasingly prominent.The drug resistant strains increased year by year has caused a lot of drugs ineffective.In order to treat diarrhea had to increase the dose of drugs.The emergence of bacterial resistance has also brought great difficulties to the treatment of infectious diseases,which leads to the increase of epidemic diseases and the increase of mortality.Plantain as natural medicinal plants is rich in resources and low in price in China.Relating to the antibiotic and chemical synthesis of drugs,Plantain containing a variety of active ingredients,targeting,low toxicity and not easy to produce drug resistance.It not only is preventing and treating the disease,but also to enhance the body's immune function,disease resistance.in addition to,Plantain has a better effect on the treatment of diarrhea.The diarrhea models were induced by castor oil in this study,observing the number of each group mice's diarrhea and calculating the loose stools rate in mice.The effect of aqueous extract of Plantago asiatica L.based on intestinal propulsive motility of mice that was observed by the movement rate of ink in the intestine.The results of this study show that aqueous extract of Plantago asiatica L.can effectively relieve diarrhea in mice.In this experiment,according to the principle of similar material compatibility,The antidiarrhea active site of the aqueous extract of Plantago asiatica L were isolated by extraction.Then,the antidiarrhea activity of the different extraction parts were determined by using of castor oil induced diarrhea,intestinal peristalsis and abdominal cavity effusion model.The results showed that the ethyl acetate part and the residual part had antidiarrhea effect and the antidiarrhea effect of them were higher than that of the aqueous extract.In addition,the ethyl acetate fraction had the strongest antidiarrhea activity.Petroleum ether,chloroform and n-butanol parts not only do not have antidiarrhea effect,but also has the role of promoting diarrhea.In the present study,the metabolites of the ethyl acetate part and the residual part were studied.Through the method of chemical identification,it was found that the ethyl acetate fraction contained only flavonoids and tannins and the residual part contained only sugar.Then,the flavonoids and tannins of ethyl acetate,and the remaining parts of sugar were determined by UV spectrophotometer.The results showed that the flavonoids content was 59.4%,tannin content was 32.9%;the sugar content was 58.1%.Finally,the experiment of ethyl acetate by liquid-liquid phase separation and composition analysis,mass spectrometry results,mainly containing apiolin-7-o-glocoside,Hyperoside,baicalin,hispidulin,5nepitrin,homoplantaginin,acteoside and other components in the ethyl acetate part and the acteoside content is far higher than that of other ingredients.In summary,the aqueous extract of Plantago asiatica L.has antidiarrhea effect in castor oil induced diarrhea.Its antidiarrhea active sites are the ethyl acetate and the remaining parts and its effective metabolites are flavonoids,tannins and sugars.the ethyl acetate fraction antidiarrhea effective substances may be one or more joint role of apiolin-7-o-glocoside,Hyperoside,baicalin,hispidulin,5nepitrin,homoplantaginin,acteoside.This study lays the foundation for the optimization of the process,the establishment of quality standard and clinical application.
