| Cartilage is a transparent and elastic connective tissue formed by chondrocytes and extracellular matrix.Extracellular matrix consists of water,polysaccharides and dense collagen and other components,which do not have blood vessels,nerves and lymphatic vessels.Chondrocytes are dispersed in which the matrix is continuously synthesized and degraded,the matrix protects the cells,providing nutrients to the cells,and the two are interdependent and maintain the stability of the joint structure.Because of the special cartilage tissue structure,when the articular cartilage damage need to be repaired,the biggest difficulty is that it is the lack of repair itself,the lack of defects necessary for undifferentiated cells,and undifferentiated stromal cells Cannot enter the damaged parts through the blood supply,similar to other organizations to repair healing.And chondrocytes embedded in the matrix,due to the density of collagen-polysaccharide,and thus the cell proliferation and migration capacity is affected,so the organization self-repair capacity is very limited.Minimal articular cartilage trauma If you do not take the intervention means to intervene,the initial small area of localized injury will gradually expand,further deterioration may even lead to osteoarthritis.In view of the previous test results are used in different study groups,different models,some of the treatment of cartilage injury,the lack of a unified,systematic assessment.In this study,we established the method of repairing the main components by repairing the cartilage injury in the shallow part,comparing the self-healing,medication and drilling methods.In this study,18 New Zealand white rabbits were used,weighing about 2.5 kg.The cartilage injury model of articular cartilage was prepared and divided into three experimental groups:the self-healing group,the drug group and the drilling group,and the effect of different treatment methods were compared.Each group of 6,and then further divided into 1 week,8 weeks two time points.The treatment group was injected with 1%sodium hyaluronate 0.2 mL per week,and the drilling group drilled three holes evenly in the middle of the defect area.The evaluation criteria for the repair of cartilage in the shallow part of the articular cartilage were composed of three parts:the behavioral score of the animal movement was combined with the Lequesne MG standard;pathology HE staining,safranine-green staining;RT-PCR method was used to detect the expression of related cytokines during cartilage formation,including Runt-related transcription factor-2(RUNX2),X type collagen(Col-Ⅹ),type Ⅰ collagen(COL-Ⅰ),type Ⅱ collagen(Col-Ⅱ),proteoglycan(Aggrecan).Comprehensive analysis,comparison of three methods of cartilage injury repair effect.Results show that after 8 weeks experimental period,using the self-healing,the treatment of drug injection,drilling shallow part of the cartilage damage,restore the tissue defect were generated,which amount RUNX2 expression in the experimental group were higher than blank group level,since the difference between the group and the treatment group was not significant(P>0.05),drilling and the extremely significant difference(P<0.01);X collagen type are below normal,and the blank group is extremely significant difference-(P<0.01);Self-healing group with drug use is extremely significant difference compared with the drilling group(P<0.01);Medication group with drilling the difference is not obvious(P>0.05);Ⅰcollagen type 8 weeks,the self-healing group significantly higher than the blank group,and the treatment group and the drilling group difference was significant(P<0.01);The two is lower than the blank group,no significant difference between groups(P>0.05);Collagen type Ⅱ blank group at 8 weeks respectively compared with self-healing group and drilling,were extremely significant difference(P<0.05);Compared with the observed group,no significant difference(P>0.05);The significant difference between groups(P<0.05);Self-healing and drilling group expression of proteoglycan content below blank group,compared with the first week is on the decline,but compared with the blank group,no significant difference(P>0.05);Medication expression quantity is still higher than that of blank group and increased slightly,and the difference between the group and the treatment group significantly(P<0.01).Prove the three methods play a repair effect.But different treatment method for defect parts repair and restoration of there exist certain differences,self-healing group of type I collagen formation of granulation tissue as the main repair;Treatment group for type ii collagen,such as transparent cartilage repair composition is closer to normal cartilage tissue;Drilling group to fibrocartilage,transparent cartilage and the mixture of calcified cartilage tissue. |
