Sodium usnic acid has a series of functions such as antisepsis, antiinflammatory, anticancer,antitumor, antioxidation, antibiosis, antiviral and antipyretic and analgesic effects, and sodium usnic acid also has positive effects on skin wound healing. Although previous studies have researched the influence of sodium usnic acid on skin wound healing, but lack of the related mechanism research.Therefore, this study use Wistar rates as experimental animals to investigate the specific role of sodium usnic acid on skin wound healing and probe into the related mechanisms. Furthermore, we use canines as experimental animals to validate the promotion of sodium usnic acid on skin wound healing.In this study, 60 maleWistar rats were randomly divided into 4 groups as negative control group,solvent control group, Gentamicin positive control group and experimental group. And the influence of sodium usnic acid to wound healing process and inflammation, proliferation phases were explored by wound healing rate experiment, pathological section observation experiment and immunohistochemical staining was used to semi quantitative analyse the content of VEGF and αSMA.And 3 male canines were randomly divided into 3 groups to validate the promotion of sodium usnic acid on skin wound healing.Results:The result of wound healing experimental in rats showed that:it is in day 3 that wound healing rates showed significant higher in Gentamicin positive control group and experimental group than that in negative control group and solvent control group(P<0.05).And the significant differences continue to day7, 10 and 14(P<0.05). In addition, the re-epithelialization time in Gentamicin positive control group and experimental group also showed significant faster than in negative control group and solvent control group(P<0.05).Pathological sections of wound in rats showed that the appearance of salient features in all phases are earlier in Gentamicin positive control group and experimental group than that in negative control group and solvent control group. And histological scores of wound on day 3,7,14 showed significantly higher in Gentamicin positive control group and experimental group than that in negative control group and solvent control group(P<0.05).The results of immunohistochemistry Experimental in rats showed that:there are Significant differences between Gentamicin positive control group, experimental group and negative control group,solvent control group in IOD of VEGF and αSMA on day 3. However, immunohistoche-mistry result of EGF did not showed any differences in four groups.The results of wound healing experimental in canines showed that: the re-epithelialization time are obviously faster in Gentamicin positive control group and experimental group than that in negative control group.The results showed that sodium usnic acid can promote wound healing in rats through shorten the inflammation period and increaseed the level of VEGF and αSMA in wound area in proliferation period.Moreover, sodium usnic acid can promote skin wound healing in canines as well.
