In recent years,scientists have succeeded in cultivating a large number o f insect-resistant,disease-resistant,herbicide-tolerant and new crop germplasms with high yield and high quality through biotechnological transformation,which has greatly enriched the crop breeding resources.Generally,after a transgenic material is obtained,a series of genetic homozygote identification,genetic stability research,agronomic trait selection,and species maintenance during industrialization are also required for its offspring.Eventually,new transgenic varieties that meet the needs of agricultural production are bred.In order to increase the expression of exogenous genes in transgenic maize and meet the industrial applications needs,a variety of plant expression vectors were constructed to transform maize.In the former research,the Bt cry1 Ah gene was transferred into maize inbred lines 31 and obtained through population screening for the Asian corn borer high-resistance material HGK60.And through agrobacterium transformation transformed hybrid lines Hi II,we obtained successful transformation event named as NK transgenic insect-resistant maize.In this study,the genetic stability and agronomic traits of exogenous genes of HGK60 inbred lines and their hybrids were analyzed for three consecutive generations,and NK materials were scre ened for three consecutive generations in order to obtain insect-resistant events with brilliant traits.Two different transgenic varieties were analyzed for genetic stability analysis through DNA(PCR detection,Southern hybridization),transcriptional level(q RT-PCR)and translational level(ELISA,test strip detection).The results showed that the transgenic cry1 Ah gene could be normally stably inherited and expressed on the normal level.The expression levels of different tissues in the same generation were different,and the protein expression levels of the same tissue in different generations were not significantly different.The insecticidal effects of HGK60 and NK corn materials were assessed by the detection of the bioactivity of the Asian corn borer both indoors and in the field.The results confirmed that the indoor bioassay of HGK60 and its hybrids had a 100% insecticidal rate against corn borer for 3 days,a field resistance test for high resistance to corn borer for 3 weeks,and a NK material for 100% of corn borer for 3 days.Insecticide rate,field resistance test for corn borer resistance level for 3 weeks is also high resistance.The agronomic traits of the transgenic insect-resistant maize were analyzed.Compared with the recipient control,the germination rate,growth period,number of ear rows,ear length,thousand grain weight and other agronomic traits of the HGK60 insect-resistant corn had no significant difference.Through many years of field trials and molecular tests,it has been proved that HGK60 transgenic insect-resistant corn has a good industrial application prospect,and NK material is an excellent reserve material for application in corn breeding for insect resistance.
