With the rapid development of biotechnology, genetic engineering has been widely used for cropbreeding, and people have obtained many breakthrough products. After obtaining transgenic crops withnew properties using genetic engineering methods, people should detect the genetic stability of theforeign gene in the transgenic plants using various molecular detection methods, and then improve thecrop species using conventionality breeding means, thereby realize the transgenic plants turn towardsproducts.This research studies the materials of the high generation insect-resistant cry1Ah maizetransformed by pollen tube pathway (PXH1and PXH2) and insect-resistant cry1Ah maize by gene gun(HF). Series of molecular detection methods plus field resistance identification have been served tostudy the genetic stability. Using backcross transformation method to get transgenic homozygous withthe excellent inbred lines. Results were as follows:(1) The cry1Ah gene can be stable heredity by generations in transgenic maize PXH1and PXH2,and reaching homozygous in T11generation. Through Southern blot analysis we proofed that thecry1Ah gene has been integrated into the transgenic maize PXH1and PXH2, and respectively in theform of single copy and dual copy.(2) The cry1Ah gene can be stable heredity by generations in transgenic maize HF, and the positiveratio reached90.75%.(3) Using backcross means we respectively obtained backcross F1generation (PXH1and PXH2)and BC2F1generation (HF) seeds. By detecting the quantity of the Cry1Ah protein expression and thefield resistance identification analyses, we found that two indexes were higher than the donor parents.(4) In the desert conditions, the survival competitiveness of transgenic maize was much weakerthan the weeds, therefore the transgenic maize would never effect the biological diversity of theenvironment.
