In order to detect and identify the ploidy differences,colchicine and pendimethalin were used to induce polyploid induction.Study in order to determine whether the higher the level of ploidy is,the better quality of Anoectochilus roxburghii has,the plant more sturdy,and the stronger the resistance,which resulted in more excellent varieties of A.roxburghii.In addition,the study used hybrid technology to obtain excellent traits from parents,combined with the excellent characteristics of the narrow-leaf A.roxburghii and the broad-leaf A.roxburghii,to create new varieties with high active components and strong disease resistance.In this study,polyploid induction was carried out with colchicine and pendimethalin at different concentrations.The optimum mutagenic conditions of colchicine were as follows:concentration of 700 mg·L-1,induction time of 24 h,survival rate of 50%,and mutation rate of43.3%.The optimum mutagenic conditions of pendimethalin were as follows:concentration of 90mg·L-1,induction time of 48 h,survival rate of 56.7%,and mutation rate of 50%.The polyploid plants obtained were more obvious than those of the control plants.The polyploid plants grew very slowly.The experiment also determined by flow cytometry to determine the level of ploidy there are significant differences.The results showed that the pollen tube growth could reach the embryo sac to complete double fertilization,and then the pollen tube and the pollen tube were transferred to the embryo.It was suggested that the pre-fertilization obstacle was not the main factor of the hybridization obstacle of the narrow-leaf A.roxburghii and the broad-leaf A.roxburghii.The abnormal embryo development after hybridization eventually led to embryo abortion,which confirmed that the post-fertilization obstacle should be the reason of low seed setting rate in the hybridization of the narrow-leaf A.roxburghii and the broad-leaf A.roxburghii.At the same time,embryo rescue was carried out in the immature embryo of the abortive embryo,and the hybrid A.roxburghii was successfully cultivated,which laid a foundation for the later research of new varieties of A.roxburghii.
