Anoectochilus roxburghii is 0rchidaceae,the Anoectochilus,herb plants,a rare traditional raw material for medicine and ornamental treature in China used for heat-clearing and detoxifying and decreasing blood pressure .The economic value and development and utilization prospect of the Anoectochilus roxburghii was important and broad . In this experiment ,we use the tissue culture seeding of Zhangzhou and Taiwan provenance of Anoectochilus roxburghii based on the method of single-factor design ,orthogonal design and quadratic regressive orthogonal design for rooting culture in photoautotrophic micropropagation technology .These tests were taken in this research by photoautotrophic incubator as follow: 1) how the different culture medium ,the different level of hormone and the different Illumination intensity, CO2 concentration and light cycle affect the rooting of Zhangzhou and Taiwan provenance of Anoectochilus roxburghii.2) Chlorophyll fluorescence analysis for the rooting process of Zhangzhou and Taiwan provenance of Anoectochilus roxburghii under the different level of hormone and different Illumination intensity, CO2 concentration and light cycle. 3)The contrast test between traditional tissue culture and Photoautotrophic micropropagation.The feasibility of photoautotrophic micropropagation was proved and made the artificial culture consummated and developed farther ,and inaugurated a new ideas and a new approach for the rapid propagation technology of Anoectochilus roxburghii .The main results were described as following :1 Through the contrast test between traditional tissue culture and Photoautotrophic micropropagation we found that, Zhangzhou provenance of Anoectochilus roxburghii and Taiwan provenance of Anoectochilus roxburghii began to take root from 20 to 30 days in the traditional tissue culture ,but they began to take root from 10 to 20 days in the Photoautotrophic micropropagation contion,and the rooting time was as faster as 1 or 2 times compared with the traditional tissue culture.In the Photoautotrophic micropropagation contion, the rooting rate of Zhangzhou provenance of Anoectochilus roxburghii improved 34% compared with the traditional tissue culture without sugar and improved 7% compared with the traditional tissue culture with sugar;the rooting rate of Taiwan provenance of Anoectochilus roxburghii had a 33% higher than traditional tissue culture without sugar and 6% higher than traditional tissue culture with sugar. 2 In photoautotrophic micropropagation condition, there was significant differences in rooting result between Zhangzhou and Taiwan provenance of Anoectochilus roxburghii from different different. In river sand (the diameter of particle 0.5 mm 1.0mm) culture medium,both Zhangzhou and Taiwan provenance of Anoectochilus roxburghii have better effects than other with rooting rate of 96% .3 In photoautotrophic micropropagation condition, the best level of hormone on rooting rate of Zhangzhou provenance of Anoectochilus roxburghii was IBA0.5mg/L+NAA0.6 mg/L+MS, the best level of hormone on root weight was IBA0.5mg/L+NAA1.0 mg/L+1/4MS, the best level of hormone on average root number was IBA1.0mg/L+NAA1.0 mg/L+1/4MS, the best level of hormone on average root length was IBA1.0mg/L+NAA0.2 mg/L+1/2MS. The best level of hormone on rooting rate of Taiwan provenance of Anoectochilus roxburghii was IBA1.5mg/L+NAA0.6 mg/L+1/2MS, the best level of hormone on root weight was IBA1.5mg/L+NAA0.6 mg/L+1/2MS, the best level of hormone on average root number was IBA1.5mg/L+NAA0.6 mg/L+MS, the best level of hormone on average root length was IBA1.5mg/L+NAA0.2 mg/L+1/4MS.4 Through the variance analysis ,significance test of regression equation for each rooting index of Anoectochilus roxburghii in the different Illumination intensity, CO2 concentration and light cycle found that :at the significant levelα=0.05,the rooting rate,root weight and average root number of Zhangzhou provenance of Anoectochilus roxburghii had significant light cycle.Illumination intensity and light cycle had significant affect on the rooting rate,illumination intensity, CO2concentration had significant affect on the root weight and average number. The rooting rate of Taiwan provenance of Anoectochilus roxburghii had significant linear relationship with illumination intensity, CO2 concentration and light cycle . At the significant levelα=0.01,the regression equation of root weight and average root length of Taiwan provenance of Anoectochilus roxburghii were significant. CO2 concentration and light cycle had significant affect on the rooting rate. Illumination intensity and light cycle had significant affect on the average root length. Illumination intensity, CO2 concentration and light cycle had significant affect on the root weight .5 Through Chlorophyll fluorescence analysis we found that under the different level of hormone ,the Zhangzhou and Taiwan provenance of Anoectochilus roxburghii had different chlorophyll fluorescence parameters variational trend of F0, Fm, Fv/Fm, Fv/F0 during rooting process.May be different treatment plant photosynthetic characteristics, comsequently making plant have different variation during pre-rooting, rooting and root-elongating stage6 Through Chlorophyll fluorescence analysis we found that under the different Illumination intensity, CO2 concentration and light cycle,the Zhangzhou and Taiwan provenance of Anoectochilus roxburghii had different chlorophyll fluorescence parameters variational trend of Fv/Fm, Fv/F0 ,NPQ,Rfd during rooting process.In illumination intensity 4000lx(0standard), CO2 concentration 800ppm(0standard) and 3.826h light 1.913h dark condition(-1standard), Zhangzhou and Taiwan provenance of Anoectochilus roxburghii had strong photosynthetic capacity. In illumination intensity 5478.197lx(1standard), CO2 concentration 504.361ppm(-1standard) and 14.174h light 7.089h dark condition(1standard), Zhangzhou and Taiwan provenance of Anoectochilus roxburghii had weak photosynthetic capacity. The results account for that: Zhangzhou and Taiwan provenance of Anoectochilus roxburghii can growth much better at In illumination intensity 4000lx, CO2 concentration 800ppm and 3.826h light 1.913h dark condition7 Compared with traditional tissue culture,the Photoautotrophic micropropagation culture could reduce production cost as well as improve production rate,and the production cost of Zhangzhou provenance of Anoectochilus roxburghii reduced about 17% and that number of Taiwan provenance of Anoectochilus roxburghii was 15%.
