Puffing is one of physiological disorders prevalently developed in citrus fruits during ripening or storage which leads to low fruit quality and storage performance.Puffing mechanism still remains unknown.In this thesis,different species of citrus were chosen as the test sample,the relevancy between puffing and different species was discussed through comparing differences in peel morphology and cell structure.Then the fruits of ‘Miyagawa' and its bud mutant system,which the former is easier to puffing while the latter is resistant,were chosen to compare morphology,peel cell structure and subcellular in fruit development processes in order to initial locate the key period of fruit ‘Miyagawa' puffiness.The pell during the key period of the fruits were used as the material for RNA-seq,and the transcriptome differences between ‘Miyagawa'and its bud mutant puffiness occurrence process were analyzed.The main results were as follows:1.By comparing the morphology of fruit and the anatomical structure of peel,the results were obtained.Mandarin fruits albedos were difference between hybrid citrus,oranges and pummelos albedos.The higher the degree of puffiness is,the losser the cell structure and lower the cell density is.The used of statistical analysis method to analyze the correlation between the main factors.The results that the puffing's level was significant related with species,albedo cell density and diameter.Among the results albedo cell density and diameter had negative correlation with puffiness.2.Through comparing differences in‘Miyagawa' and its bud mutant system in morphologh and peel cell structure.Changes of albedo subcellular structure observed by transmission electron microscope.The key period of puffing happened during 58 and 88 days after flowering.3.High-throughput sequencing technique was used to analyze the albedo of 'Miyagawa' and its bud mutant system during the critical period.The different expression genes of transcriptome were mainly enriched in ‘membrane,lipid metabolism',‘biosynthetic processes',‘oxidoreductase activity' and ‘catalytic activity' etc.
