This research adopts the physiological and biochemical,molecular biology and omics technology,explores the mud skipper?Boleophthalmus pectinirostris?physiological mechanism of ammonia poisoning,and taurine and methionine alum amine?the oledata.mso?to evaluate the slow-release effect of ammonia nitrogen.This study consists of three experiments:?1?Comparative transcriptome study of differential genes under acute ammonia stress in mud skipper.In this paper,the experiment was carried out on a 96-h ammonia stress test with mud skipper?15.14+0.05?g as the experimental material,and the high-throughput sequencing was performed on the liver samples.The results showed that 87 654Unigene were assembled.By comparing the transcriptome data between different treatment groups,there were 10,149 genes showing differential expression,of which 4,727 were up-regulated,and 5,467 were down-regulated.The gene of differentially expressed genes was analyzed by the enrichment of GO function,and the enrichment of 4 121 genes was obtained.The results showed that the ammonia stress could induce the inflammatory reaction of the fish,taurine and MSO could alleviate the ammonia poisoning,but the detoxification mechanism was not the same.?2?Effects of acute ammonia nitrogen stress on the expression of immune related genes in mud skipper.The experiment was carried out on a 96-h ammonia stress test with mud skipper?15.14+0.05?g as the experimental material.The results showed that theLC500 was 8.99 mg/L.The expression of TNF and IL-6 was down-regulated after stress,and it was up-regulated at 12 h and 96 h.The expression of IL-1 and IL-8 gene was increased and then decreased.The results showed that ammonia nitrogen stress on the big play with fish inflammatory?TNF alpha,IL-1,IL-6 and IL-8?the expression of related genes have a negative impact,promoting inflammatory factor induced by continuous stimulation of cell apoptosis may be lead to big to play with the fish ammonia poisoning death.?3?The effects of taurine and MSO on the expression of anti-oxidative and inflammation-related genes in mud skipper under acute ammonia nitrogen stress.In this experiment,a total of 4 treatment groups were set up:1?control group?and intraperitone al injection of saline;2.Group 2 injection of ammonium acetate;Group 3 injection of ammonium acetate and taurine;Group 4 injection of ammonium acetate and MSO.The results showed that SOD,CAT,GSH-PX and GST gene expression were the highest in group 2,while group 3 and group 4 were the lowest.In the control group,the expression of TNF gene was significantly higher than that in other groups.The highest expression levels of IL-1 and IL-8 were found in group 4 and group 2 respectively.The results showed that ammonia stress could negatively affect the expression of anti-oxidant?SOD,CAT,GSH-PX and GST?and inflammatory?TNF-alpha,IL-1,IL-6 and IL-8?genes in mud skipper.Taurine can play a better slow-release role;MSO can prevent overproduction of reactive oxygen species,but it has no effect on inflammatory response.To sum up,this paper makes a systematic experiment and analysis on the mechanism of ammonia poisoning of mud skipper,and it is also the first time to study the transcriptase data of mud skipper.The data to get involved in inflammatory reaction and immune defense system offers a wide range of some of the new gene resources,analyze the differentially expressed genes can also help us understand the fish ammonia poisoning mechanism,also has been clear about the taurine and methionine and ammonium alum?the oledata.mso? ease fish ammonia poisoning mechanism.
