| Boleophthalmus pectinirostris is a kind of economic fish with broad market prospects.As a kind of coastal cave dwelling fish,B.pectinirostris has become a special case of fish because of its unique behavior and physiological and biochemical characteristics suitable for amphibian life.Although it inhabits in the mudflat of intertidal zone with complex environment,it has strong disease resistance.Therefore,the acquired immune mechanism of B.pectinirostris which can adapt to various adverse environments has high research value.Major histocompatibility complex(MHC)is a kind of gene encoding glycoprotein of cell surface,which plays an important role in the adaptive immune system of all teleosts.The research on MHC gene has been one of the hotspots in fish molecular immunology and disease-resistant assisted breeding.The purpose of this study was to analyze and identify the sequence of MHC Iα,MHC IIα,MHC IIβ and MHC IIγ genes of B.pectinirostris,construct phylogenetic tree to analyze the evolutionary relationship between MHC gene of B.pectinirostris and other identified fish,use absolute fluorescence quantitative technology to evaluate the difference of MHC gene expression in different tissues of healthy individuals,and use quantitative real-time PCR technology to study the expression of MHC Iα,MHC IIα,MHC IIβ and MHC IIγ genes in the liver and spleen of the main immune organs of the body after injection of Vibrio anguillarum and viroid poly(I:C)respectively under high salt stress.The results showed that MHC Iα、MHC IIα、MHC IIβ and MHC IIγ genes were expressed in different tissues,although the expression level was different.The high expression mainly occurs in liver,gill,kidney,spleen and intestine,probably because these parts are important parts of innate and adaptive immune response of fish,and also the most part of immune cells,including antigen-presenting cells.And in the face of harsh environment,the MHC gene of Boleophthalmus pectinirostris will participate in the related immunity quickly,and its expression will change relatively,which proves that the MHC gene of Boleophthalmus pectinirostris is involved in its immune expression and plays an important role. |
