There are abundant biological resources in the ocean.About 80% of the species on the earth inhabit the ocean.Marine organisms are important sources of molecular resources such as biological functional proteins and active peptides. At present, marine organisms have become a treasure trove of various functional proteins and active peptides,a large number of functional proteins(such as protamine, mussel foot silk protein, collagen and nutrient proteins)and medicinal peptides extracted from marine organisms(Such as antibacterial peptides, toxin peptides, tumor suppressor peptides and hydrolyzed peptides, etc.,have been or are becoming the leading molecules in protein and peptide engineering research.Among them,Mussel foot protein(Mfp)has strong adhesion ability and is not affected by water environment. Therefore,it has broad application prospects in many industries such as medicine and chemical industry.A new bio-adhesive with a chemical adhesive.Studies have shown that dopa plays an important role in the adhesion of the foot silk protein and intramolecular cross-linking.However,the redox system involved in the adhesion process of the mussel foot silk and its molecular mechanism are still unclear. In order to further explore the structure and function of other antioxidant proteins in the mussel foot silk,a new glycine-and cysteine-rich CG-protein was identified for the thick-shelled mussel foot.Recombinant expression and related functional studies;in addition, fish skin mucus has an extremely important physiological significance for fish survival.Boleophthalmus pectinirostris is a special bony fish that can live in amphibious life. The skin mucus is essential for the immune defense, osmotic pressure maintenance, and respiratory processes of the large-slung fish, but there is no report on the molecular composition of the skin mucus of the large-boiled fish.In order to understand the biological significance of the skin of the big bomb and its mucus in the special life habits of the big bomb, the microscopic structure of the skin of the big bomb and the antibacterial activity and proteomics of the skin mucus were carried out.The results showed that the novel foot silk antioxidant protein CG-protein has obvious antioxidant activity and has a specific distribution and complex interacting protein system in the foot and foot glands. It is speculated that this protein plays a protective role in preventing the early oxidation of the foot silk protein during the adhesion of the mussel foot silk. The above studies further clarified the existence of a variety of antioxidant active proteins in the mussel foot silk, which is of great significance for understanding the adhesion molecular mechanism of the mussel foot silk.Studies on the skin and mucus of large-bodied fish showed that the skin tissue of the large-bodied fish had a special dermal vesicle structure, and viscous polysaccharides with sialylation and sulfonation existed in the vesicles.Further analysis showed that the skin mucus of the large-elastic fish had obvious Broad-spectrum antibacterial activity;using transcriptome and proteome binding strategies, more than 500 protein molecules were identified from the skin mucus of large-elastic fish, mainly including various metabolic-related enzymes and immune-related proteins, indicating that the skin of the large-elastic fish has mucus Complex molecular composition and physiological functions. On the one hand, the above research lays a foundation for understanding the molecular structure and function of the foot silk antioxidant protein and developing a new antioxidant protein based on the mussel foot silk;on the other hand, the skin structure, skin tissue transcriptome, skin from the big bomb Obtaining systematic research data on the antibacterial activity and molecular composition of mucus, obtaining systematic research data for understanding the biological functions of fish skin and mucus, and screening and developing special active proteins and molecular markers for subsequent follow-up Foundation.
