Study On The Technique Of Making Frozen Semen Of Tibetan Sheep In Qinghai

The research of artificial insemination and frozen semen technology can promote semen production standardization and maximize the utilization efficiency of excellent Tibetan ram sheep.These technologies also reduce the feeding number of Tibetan ram sheep and the pressure of grassland which protects the ecological environment and promotes the economic benefits of Tibetan sheep industry.It is significant to the development of modern ecological organic animal husbandry in Qinghai province.This experiment was conducted to study the fine semen collection of ram and establish a technical standard system.The study had figured out the different diluents of this formula,freezing test program,water bath thawing temperature,thawing time and the semen quality index.The optimal dilution formula of frozen semen of Tibetan sheep was: 100 mL distilled water,0.5g fructose /glucose,3.7g glucose,20 mL egg yolk,5mL ethylene glycol,3.7g citric acid/sodium citrate,125 mL VB.The optimal tubule freezing temperature curve and thawing temperature and time: Frozen curve:-10--4?(cooling rate of 5?/min),-10--100?(cooling rate of55?/min),-100--140?(cooling rate of 20?/min).Thawing method: 38?(20 s)-40?(15s)water bath thawing after freezing.The sperm viability is 41.60%.Each dose of dynamic semen was 21.2million.The malformation rate of thawed sperm was 10.67%.The intact rate of thawed sperm was 84.93%.The bacterial colonies number in each dose of sperm is 14.9 has achieved DB63/T1436-2015 standard and the requirements of GB4143-2008(bovine frozen semen).The popularization and application of estrus synchronization,laparoscopic and frozen semen artificial insemination,lambing period,early weaning lamb feeding and efficient management can greatly shorten the breeding cycle of Tibetan sheep,improve the level of Tibetan sheep breeding and promote the industrialization development of Tibetan sheep.
