Semen cryopreservation frozen technique use liquid nitrogen as a frozen source, stored at ultra-low temperature after special treatment, so as to be stored for a long time. Compared with other species, ovine semen has the feature in physiological and biochemical. It has lower resistance for low-temperature, the frozen effect is not ideal till now. It is hard to promote the technology in artificial insemination of ovine.In order to improve the quality of frozen semen of ovine, we use the monitor evaluation of sperm motility, acrosome integrity, membrane integrity and aspartate aminotransferase and lactate dehydrogenase activity etc. after thaw in this experiment. And we have optimization study for ovine semen diluent formula, tubules frozen semen produce program, and the method of centrifugal for the removal of seminal plasma, results are as follows:(1) The F formulations which contains D-fructose, citric acid is the best in the six kinds of frozen semen dilution formulations, the sperm motility after thaw is0.41.(2) The best equilibrium time for the process in making the tubules semen is2.5h, as the equilibrium time prolong the quality of thaw semen is decrease. The best result of thaw program is at the temperature of40℃, for10s, the sperm motility after thaw was significantly (P<0.01) higher than the program of70℃,6s.(3) In the key link of semen frozen which is semen frozen cooling speed, frozen cooling program b is the best(The cooling speed between4℃~-15℃is5℃/min, for-15℃~-90℃is20℃/min.).(4) The best condition of centrifugation for the removal of seminal plasma is as followings:centrifugal at the speed of1250r/min for5min. The sperm motility after thaw which undergo centrifugation for the removal of seminal plasma was significantly higher (P<0.01) compare to the semen which have not undergo centrifugation except semen seminal.In summary, this study is having optimal study in sheep semen cryopreservation-freezing and thawing program, frozen dilution formula and centrifugal for the removal of seminal plasma technology. And the ovine sperm mobility after thawing is more than0.35, which can meet the need in production.
