P.amurense is the only source of plants for the traditional Chinese herbal medicine.Now the resources of wild P.amurense were seriously damaged,through the investigation found that the population of wild P.amurense self-renewal obstacles,now has become an endangered species.Because seed has deep dormancy and low germination rate,This severely limits the herbs in the tradition of expanding production.Therefore,It has important meaning to discuss factors affecting seed germination,In this paper,seed and leaves as experimental materials.Through the water permeabilityand air permeability of seed.Bioassay of extracts from different parts and bioassay of endogenous inhibitory substances,isolation and identification,To explore the characteristics of seed dormancy,and the causes of seed germination difficult.The results show that:Study of breathable absorbent and seeds of P.amurense.Completeand coat of P.amurense seed after soaking in water,large difference in waterabsorption,After 100 h,the seeds reached the saturated state;By comparing the determination of stratification time of different seed and without stratification analysis,That kind of low permeability of the shell of P.amurense,can significantly inhibit the respiration of seeds.After stratification treatment,Inhibition of seed coat on respiration,It is beneficial to breaking dormancy and promoting germination.Using water and methanol as extractant,find seed,endosperm and leaf extractcould inhibit the germination and seedling growth of Chinese Cabbage,and compared with methanol extract,water extract,the latter is more inhibitory.as well as prove Obaku seed coat,endosperm and leaf contained germination inhibitory substance.It is also proved that the germination inhibitor is more soluble in methanol,It also provides a theoretical basis for better selection of leaching solution.Using petroleum ether,ethyl ether,ethyl acetate,methanol and water as extracting agent get the corresponding extract of P.amurense seed,endosperm and leaf,found the germination percentage of each organic cabbage seed?the growth of root length and seedling height were inhibited in different degrees.The inhibition of extract from different parts was leaf > endosperm > seed coat,The inhibitory effect of different organic solvents on the extraction of methanol was the highest,diethyl ether,methanol,ethyl acetate and water extracts had a weak inhibition.The seed of endogenous inhibitory substances were isolated and biological activity determination use systematic solvent separation,By petroleum ether,ethyl ether,ethyl acetate and methanol as extractant get the corresponding extract of P.amurense seed,endosperm and leaf.Use GC-MS analysis of organic phase and identify the types of substances present.Most of the organic compounds identified from the seed coat,endosperm and leaves of the ether phase are acids,esters and alcohols,the growth of cabbage seeds has an inhibitory effect.
