The expression of Sirt2 and Sirt3 genes in porcine visceral tissues,SNPs and their association with meat quality traits of pig were studied in this study,the relationship between sirt2 and sirt3 genes and meat quality traits of pig was explored.The results were as follows.(1)Expression of sirt2 and sirt3 genes in porcine visceral tissuesThe expressions of two genes in the heart,liver,spleen,lung,kidney,pancreas,hind leg muscle and longissimus dorsi were detected and analyzed by used the cDNA sequence of two genes provided by GenBank.The gene sequence,protein sequence and specific gene expression were also explored,the results were as follows.There were extremely significant differences(P<0.01)in sirt2 gene expression of heart and hind leg muscle in Xiangcun black pig,and significant difference(P<0.05)in lung,pancreas and kidney;while in Daweizi pig,significant difference were found in the lung,heart,liver and hind leg muscle(P<0.05).The gene expression analysis of the same tissue in Xiangcun black pig and Daweizi pig shows that significant difference(P<0.05)was found in the pancreas,and extremely significant difference(P<0.01)in liver.There were extremely significant difference(P<0.01)in sirt3 gene expression of heart and lung in Xiangcun black pig,and significant difference(P<0.05)in lung,spleen and longissimus dorsi in Daweizi pig,The gene expression analysis of the same tissue in Xiangcun black pig and Daweizi pig showed that there were significant difference(P<0.05)in lung and spleen.(2)Gene mutation site in SNPs and association analysis of meat quality traitsThe mutations of sirt2 and sirt3 genes were detected and analyzed by primer design,PCR amplification,sequence and restriction enzyme digestion.The meat quality traits of Xiangcun black pig and Daweizi pig were associational analyzed to explore whether there was significant correlation between two genes' mutations and meat quality traits.The association analysis of two swine breeds in meat quality traits: The sirt2 gene had a C/T mutation site on the exon 8,formed three genotypes of CC,CT and TT.There was significant difference(P<0.05)between muscle color value,water loss rate and intramuscular fat content in Daweizi pig,and there was significant difference(P<0.05)intramuscular fat,drip loss and water loss rate in Xiangcun black pig;The sirt3 gene had a A/G mutation site on the exon 5,formed three genotypes of AA,AG and GG,there was significant difference(P<0.05)correlated with drip loss,water loss rate and intramuscular fat content in Daweizi pig.Only significant correlation(P<0.05)was found in the muscle color value in Xiangcun black pig.As the related genes of meat quality traits,the study of Sirt2 and Sirt3 genes may provide theoretical support for the improvement of pork quality,and accumulate experience for the utilization of local pig breeds.
