| It is great significance to achieve the effective preservation of high-quality Lilium resources and improve the survival rate of tissue culture seedlings of Lilium,which make sence in the innovation of Lilium germplasm and promoting the industry development.At present,there are few species of Lilium that are conserved in vitro using the restricted growth method,and the state of Lilium growth during preservation is still not clear.This study explored the effects of different sucrose concentrations on the in vitro conservation of Lilium,identified the commonness and individuality of in vitro conservation of different species of Lilium,and established a preliminary technical system for the acclimatization and transplanting of three varieties of cut-flower Lilium,and improved the survival rate.,which laid the foundation for the industrialization of Lilium and the identification of transgenic and polyploid plants.The main findings are as follows:1.Optimum sucrose concentration screening for different kinds of Lilium in vitro preservationLilium tissue culture seedlings with 0.5 cm diameter bulbs were stored in medium supplemented with different sucrose concentrations to investigate the in vitro preservation characteristics of different species of Lilium.The results showed that the optimum sucrose concentration of L.longiflorum Thunb.hybrids’White Heaven’,OT Lilium hybrids’Yelloween’and’Red Dutch’,L.distichum Nakai and L.martagon var.pilosiusculum preservation mediums was 60 g·L-1,in the range of 30 g·L-1 to 60 g·L-1,by increasing the sucrose concentration in the medium,the growth rate of Lilium leaves can be significantly inhibited,the diameter of bulbs can be significantly increased,the root system can be developed,which can be preservated separately for 92 d,91 d,91 d,91 d and 90 d.The optimum sucrose concentration of Oriental hybrids’Tiger Woods’and’Double Suprise’and L.dauricum Ker-Gawl.preservation mediums was 90 g·L-1,in the range of 60 g·L-1 to 90 g·L-1,by increasing the concentration of sucrose in the culture medium can significantly inhibit the growth rate of Lilium leaves,significantly increase the bulb volume,and strengthen the roots,the in vitro preservation time can be extended to 122 d,109 d and 90 d,respectively.2.Establishing a preliminary technology system for acclimatization and transplanting of three cut-flower varieties LiliumSelection of Lilium seedlings:Lilium that were subcultured twice and had strong roots and leaves were the most suitable for transplanting.The survival rate of the three cut flowers Lilium can reach more than 88.0%.Acclimatization time:Lilium tissue culture seedlings were opened at 25±1°C,and transplanted after acclimatization at different times.The experimental results showed that the best acclimatization time of the L.longiflorum Thunb.hybrids"White Heaven"and Oriental hybrids"Double Suprise"and’Tiger Woods’seedlings was 5 days,and OT Lilium hybrids’Yelloween’and’Red Dutch’was 3 days.After transplanting,the original leaves of the tissue culture seedlings can maintain normal growth,and the survival rate of transplanting can reach more than 86.6%.Transplanting time:The well-grown Lilium plantlets with intact roots and leaves were selected and transplanted at different times after acclimatization.The results showed that the best transplanting time for the L.longiflorum Thunb.hybrids"White Heaven"and OT Lilium hybrids’Yelloween’and’Red Dutch’tissue culture seedlings was April,May,June,September and October,and the survival rate of transplanting could reach more than 90.0%.The best time for transplanting of Oriental hybrids’Tiger Woods’and’Double Suprise’was April,May,September and October,and the survival rate of transplanting could reach more than 80.0%... |
