Sphallerocarpus gracilis (SG), which has a high nutritional and medicinal valuedeeply, is a perennially natural herb and pure natural green health protective food. Inthis paper, SG, as raw material, produced from Shandan, and from which theextraction process parameters of Sphallerocarpus gracilis polysaccharides, thechemical structure and its hypoglycemic effect were studied.Three extraction method: conventional hot water extraction, ultrasonic assistedextraction and ultrasonic combined enzymatic extraction were used to extractSphallerocarpus gracilis polysaccharides. The optimum conditions of extraction weregained by single factor experiment and response surface optimization. Yield ofSphallerocarpus gracilis polysaccharides as evaluation, through comparing threeextraction methods, the ultrasonic combined enzymatic extraction method condition:0.4%papain,2%cellulase enzyme, reaction time:25min, reaction temperature:61?,PH5.8, solid-liquid ratio1:40(g/mL), ultrasonic power61%is the most suitableextraction data. The Sphallerocarpus gracilis polysaccharide yield was19.01%Sphallerocarpus gracilis polysaccharide (SGPHWE) was deproteinized anddialysised by enzyme-linked usage+sevage, DEAE-celloluse52columnchromatography to obtain high purity Sphallerocarpus gracilis polysaccharide, whichcontent is93.27%. The results of GC-MS analysis showed that: Sphallerocarpusgracilis polysaccharide is composed of five monosaccharides: Rha, Ara, Man, Glu andGal, whose molar ratio number is:4.12:8.99:5.45:65.94:15.5.Molecular weight ofSphallerocarpus gracilis polysaccharide is measured by GPC: Mw=7.413×105,Mn=2.104×105g/mol.By examining the Sphallerocarpus gracilis polysaccharide onscavenging capacity of DPPH radicals, hydroxyl radicals and superoxide anion radical,ferrous ion chelation and reducing power, the results showed that SGP has obviousantioxidant activity.By using Sphallerocarpus gracilis polysaccharide, Sphallerocarpus gracilispolysaccharide sulfate (SSGP), which was synthesized by CSA/Py, sulfur content(S%) in SSGP as evaluation, the preparation conditions of SSGP was established:esterification reagent ratio(chlorine acid:pyridine) number was1:1.3, reaction timewas3.4h, reaction temperature was65?, sulfur content of SSGP was up to2.08%. The structure of SSGP were characterized by IR, GC-MS and other methods, theantioxidant activity was evalued. It is noted that the monosaccharide species were notchanged after sulfation, but the relative content of monosaccharides was changedobviously. The antioxidant capacity of sulfated Sphallerocarpus gracilispolysaccharide has been significantly improved than before.Diabetic rat model was induced by alloxan (ALX, alloxan) with different dosesof SGP gavaged diabetes (diabetes mellitus, DM) rats. After the expiration of theexperiment, it was noted that SGP could significantly reduce blood glucose levels ofalloxan induced diabetic rats, reducing pancreatic glucagon levels and serumcholesterol (TC)?triglyceride (TG) and low density lipoprotein (LDT-C)of diabeticrats, increasing serum insulin levels and the levels high-density lipoprotein (HDT-C)of diabetic rats; To a certain extent, it also could repaire the damage of alloxan to liverand kidney of diabetic rat, and significantly reduce the content of alanineaminotransferase (GPT), aspartate transaminase (GOT), bilirubin (TBIL), creatinine(BUN) and blood urea nitrogen (Scr) in serum of diabetic rats, being able to improveantioxidant function, increased SOD and CAT activities of serum, liver and kidney indiabetic rats, reducing MDA levels, eliminating free radicals, reducing damage of freeradical to the body and islet cells, promoting secretion of insulin, decreasing the levelsof serum glucose.Polysaccharides was extracted from Sphallerocarpus gracilis by using ultrasonicand combined enzyme method in this research, optimized the technology of extractionof Sphallerocarpus gracilis, of which determining the molecular weight, thecomposition of monosaccharide, and the antioxidant activity in vitro; SSGP wasproduced by SG, which was the raw materal, considering the condition of technicsof SSGP, and from which assaying the molecular weight, composition ofmonosaccharide and the antioxidant activity in vitro; By establishing the diabetic ratmodel with four oxygen pyrimidine and lavaging the different doses of SG, it turnedthat SGP is a natural plant polymer antidiabetic products, with non-toxic side effects.It could be exploited into a new antidiabetic drugs through further researches.
