| Objective To investigate the expression levels of miR-21、miR-25、 miR-132and miR-221in polycystic ovary syndrome (PCOS)patients with hyperandrogenism(HA) group、non-HA PCOS patients group and control group, in order to explore the role of microRNA in the pathogenesis of PCOS patients HA.Methods A total of81infertile patients who undergoing in vitro fertilization-embryo transfer/intracytoplasmic sperm injection (IVF/ICSI-ET) from September,2013to February,2014at reproductive medicine center of Xiangya hospital were collected.They included23PCOS patients with HA,27non-HA PCOS patients and31non-PCOS patients. Follicular granulose-luteal cells were obtained after oocyte retrieval from the81patients.The expression of miR-21、miR-25、 miR-132and miR-221were determined by qRT-RCR from the Granulosa cell after24-48h in vitro culture. Spearman’s analysis were used to evaluate the relationship between the levels of these microRNAs and FAI (free androgen index). The roles of these microRNAs on the pathogenesis of PCOS with HA were analysed. Results1. The granulosa cells attached on the culture medium after24hours in vitro culture and multiple interconnections between the cells as typical granulosa cells after cultured48hours in vitro.2. The expression of miR-21and miR-25in granulosa cells of Group A were significantly lower than that of the other two groups(P<0.05).3.miR-21and miR-25were negatively associated with FAI(r=-0.331,P=0.003; r=-0.285,P=0.01).Conclusion The lower expression of miR-21and miR-25may related to the pathogenesis of PCOS patients with HA. |
