78%of the total micro amount of human.The species of bacteria in intestinal is between 400 to 500,which can be divided into origin flora and foreign flora,most of the normal flora in intestinal is foreign flora.In addition to bacteria,human body also has the normal virus group?normal fungus group?normal helix group which have their physiological function.Human being had built a symbiotic relationship with the normal bacterium group in the long evolution process,which play a important on promoting the perfection of human body physiology and especially the mature of the immune function.Normal flora has formed interdependence?mutual benefit?mutual coordination and restraining with human body which reflect the dynamic balance of human micro ecological,when it has balance can lead to health,otherwise is pathogenic.Stabilization is one of the most notable features of intestinal flora.This research can provide a certain basis for the evaluation on Ultrafine Granular Powder of Herbal Medicine efficacy and safety through the comparison and analysis of the influence on intestinal flora and gastrointestinal tissue after dosing Rhodiolae rosea and Salvia miltiorrhiza Ultrafine Granular Powder of Herbal Medicine.traditional decoction powder and traditional decoction.Method1.Mice were randomly divided into the normal group?each dose of Ultrafine Granular Powder of Herbal Medicine group?traditional pieces group and the traditional pieces powder group Bifidobacterium?Lactobacillus?Escherichia coil and Enterococci were selectively cultured and counted after 14 days' administration intragastrically to compare the change before and after administration the bacterium were cultured in the corresponding medium after diluting and homogenizing the content.2.The stomach and ileocecal organization which fixed by 10%fromaldehyde and HE staining in mice were taken to observe the pathological section.3.Mice were randomly divided into the normal group?each dose of cell-broken pieces group the conventional decoration pieces group and the traditional powder group.After the last administration,the dung genome extraction kit was used to extract the total DNA in the ileocecal stool.The total DNA was amplified by PCR using bacterial 16S rDNA which was used to analyze the bacterial fingerprinting by using DGGE,that we would compare the change of intestinal bacterial populations in mice before and after administration.Result1.The change of intestinal pathogenic bacteria and probiotic bacteriaRhodiolae rose:(1)The Escherichia coli and Enterococcus of each Rhodiolae in mice intestinal have no difference when compare with the normal group.(2)Compare each group's laetobaceerillus with the normal group,the traditional pieces group and traditional pieces powder quantity significantly reduced and meaningful difference,each cell-broken pieces had no significant difference.(3)Compare each group's Bifidobacterium with each other,1/8 Ultrafine Granular Powder of Herbal Medicine has most but has no significant difference with the normal group,traditional pieces' quantity reduced and meaningful difference with the normal group.Compared with the traditional pieces,each Ultrafine Granular Powder of Herbal Medicine quantity increased and the 1/8 Ultrafine Granular Powder of Herbal Medicine group had the meaningful difference.Compared with the traditional pieces,1/4?1/8 Ultrafine Granular Powder of Herbal Medicine quantity increased and had the meaningful difference.Salvia miltiorrhiza:Compared with the normal group,Bifidobacterium and laetobaceerillus in 1/8 Ultrafine Granular Powder of Herbal Medicine group and isodose Ultrafine Granular Powder of Herbal Medicine group were higher and meaningful difference,the Escherichia coli and Enterococcus of each Rhodiolae in 1/8 Ultrafine Granular Powder of Herbal Medicine group and isodose cell-group pieces group have no difference.(2)Compared with traditional pieces,1/8 Ultrafine Granular Powder of Herbal Medicine group's Bifidobacterium increased and had the meaningful difference(P<0.01),isodose Ultrafine Granular Powder of Herbal Medicine group's Bifidobacterium increased and had the meaningful difference(P<0.05).(3)The fouo bacteria in traditional pieces and traditional pieces powder group had no difference with the normal group.1.Gastrointestinal pathology resultThe results of Rhodiolae rose of blank group's mice stomach ileum pathology showed:the structure is complete,each layer of mucosa?submucosa?muscular layer and serous lager are intact,the special structure of basic integrity in every layer have no obvious hyperemia edema?inflammatory cell infiltration?The other medicine groups all have some inflammatory cell infiltration and vascular lesions of expansion hyperemia in the stomach and ileum,1/8 Ultrafine Granular Powder of Herbal Medicine group is the worst when isodose Ultrafine Granular Powder of Herbal Medicine is similar to Conventional group and Traditional powder group.The results of Salvia miltiorrhiza mice stomach ileum pathology showed:the structure is complete,each layer of mucosa?submucosa?muscular layer and serous lager are intact,the special structure of basic integrity in every layer have no obvious hyperemia edema?inflammatory cell infiltration.2.PCR-DGGE fingerprinting resultBacterial fingerprinting of Rhodiolae rosea show:shanno-wiener index and species richness of each administration group are highest and has the highly significant difference compared with normal group(P<0.01),meaningful difference when compared with conventional pieces group and traditional powder group(P<0.05).1/4 Ultrafine Granular Powder of Herbal Medicine group has the second high Shanno-Wiener index and species richness and has significant difference when compared with normal group(P<0.05).Each group has a certain similarity with the normal group(30%-45%),the similarity of isodose Ultrafine Granular Powder of Herbal Medicine group is highest(64.05%),1/2 and 1/4 Ultrafine Granular Powder of Herbal Medicine are second(61.68%),the other groups similarity are between 30%to 60%.Clustering analysis showed that the bacterium in mice of every group can be clustered into group basily and have the cross connection between group part.The Shanno-Wiener index and species richness of each Salvia miltionhiza administration group are highest group,traditional powder group is the highest Similarity with normal group(0.50),the second is 1/8 cell-broken pieces group and isodose cell-broken pieces group,the similarity of other groups are between 0.2-0.4.Clustering analysis showed that bacterium in mice of every group can be clustered into group basily and have the cross connection between group part.ConclusionThe Rhodiolae rosea and Salvia miltiorrhiza Ultrafine Granular Powder of Herbal Medicine have no harmful effect on intestinal flora in mice and gastrointestinal tissue,but can adjust the intestinal flora under using reasonably.The Rhodiolae rosea Ultrafine Granular Powder of Herbal Medicine and traditional pieces?traditional pieces powder have damage on gastrointestinal tissue,the Salvia miltiorrhiza Ultrafine Granular Powder of Herbal Medicine and traditional pieces?traditional pieces powder have no damage on gastrointestinal tissue.
