ObjectivesThe therapeutic effects of B2013-PA on ODM rats will be researched and it will provide the appropriate basis for the clinical application of B2013-PA.Methods1.Male SD rats weaned 1-3 days were fed with high fat diet for 6 weeks.After changed ordinary drinking water to syrup(10%fructose solution),fed the rats for 7 weeks with high fat diet to set up DIO model.The DIO rats were injected STZ through tail vein.The ODM rats model were confirmed by continuous observation of weight and measurement of OGTT.2.The qualified ODM rats were divided by stochastic equilibrium method according to fasting glucose(or OGTT results)and weight into 11 groups,such as:Ordinary group,DIO group,ODM group,sibutramine group,metformin group,B2013-A 0.1 mg/kg group,B2013-PA 0.025,0.05,0.1 or 0.2mg/kg group and so on.3.There were several indicators to be measured,such as:serum insulin,blood glucose value,body and feeding weight,liver biopsy,weight of right hind leg tricep,gonads around fat,perirenal fat and subcutaneous fat of rats and so on.4.The data was compared between groups by t test statistics.Results1.The highest blood glucose value of STZ 25 mg/kg of DIO rats was more than 16.7 mmol/L.The 2 h blood glucose value of STZ 25 mg/kg of DIO rats was more than 11.lmmol/L.The DIO rats were injected STZ 25 mg/kg through tail vein,that prepared the ODM rats model efficiently and stably.2.B2013-PA and B2013-A inhibited the appetite of ODM rat.ODM+metformin group was the best;its effect was reflected in the whole process of administration.The effect of ODM+PA and ODM+B2013-A groups was reflected in the first week.3.Blood triglyceride:That compared with the ODM+ excipient group,ODM+metformin group,ODM+sibutramine group and ODM+PA 0.025 mg/kg group reduced blood triglyceride.They had significant difference.The effect of B2013-PA 0.025 mg/kg was worse than metformin.4.Fasting blood sugar levels:That compared with the ODM+excipient group,ODM+metformin,ODM+sibutramine,ODM+PA 0.025 mg/kg and ODM+PA 0.1 mg/kg groups reduced fasting blood sugar levels.They had significant difference,but the effect of sibutramine and metformin was stronger than B2013-PA.5.Random blood glucose levels:That B2013-A and B2013-PA can reduce random blood glucose levels of ODM rats,but the effect of metformin and sibutramine is the best.6.B2013-PA 0.025 mg/kg significantly reduced serum insulin concentration,increased insulin sensitivity index and reduced insulin resistance index of ODM rats.The effect of Metformin is strongest than B2013-PA 0.025 mg/kg and sibutramine.B2013-A is worse than B2013-PA.7.Compared with the ODM+excipient group,PA 0.025 mg/kg,PA 0.05 mg/kg and PA 0.2 mg/kg improved the degree of fatty liver.It had significant differences,but Sibutramine and metformin had no effect.B2013-PA effectively improved the degree of liver steatosis.8.Compared with the ODM+excipient group,PA 0.1 mg/kg and Sibutramine reduced abdominal circumference.It had significant differences.ConclusionsThe results show the treatment of B2013-PA for ODM rats,that this drug can lose weight,suppress appetite,reduce blood triglyceride levels,improve insulin sensitivity index and insulin resistance index,improve the degree of fatty liver,reduce the random blood glucose and fasting blood glucose.It has a certain effect on the ODM rats.
