Objective:To study the change of retinol binding protein 4(RBP4) and visfatin concentration in pilot's serum, visfatin genotype and allele distribution, and to investigate the relation of serum retinol binding protein 4 and visfatin gene phenotype changes in pilots, coronary heart disease patients and healthy persons.Methods:Serum sample were taken from 477 military pilots(pilot group),44 patients with coronary heart disease(CHD group) and 202 health ground service personnel (control group). Pilot group and normal healthy people respectively divided by age as 21-30,31-40 and ?41 years sub-groups. Pilots were also grouped into<1000 h, 1001-2000 h,2001-3000 h and ?3001 h sub-groups by flying hours. The serum level of RBP4 and visfatin is measured by enzyme linked immune sorbent assay(ELISA) meanwhile, polymerase chain reaction-direct sequencing (PCR-sequencing) to detect visfatin-1535C>T polymorphism genotype and allele frequencies, measurement of height, weight, blood pressure, and collect and record the appropriate biochemical data:glucose (FBG), total cholesterol (TC), triglyceride (TG), the application of statistical software for statistical testing.Results:1.The serum level of RBP4 and visfatin concentrations in CHD group were (322.73±132.55)pg/mL?(6.10±2.18)ng/mL, in pilot group were(259.15± 69.23)pg/mL?(4.86±1.50)ng/mL, in control group were (225.48±88.42)pg/mL (4.39±1.62)ng/mL. The comparison between the three groups showed significant difference(P<0.05), and the CHD group compared with pilot group and control group that the serum level of RBP4 and visfatin in CHD were significantly higher than control group (P<0.05), the pilot group compared with control group that the serum level of RBP4 and visfatin in pilot were significantly higher than control group (P<0.05); 2. In the same age group, the RBP4 concentration in pilots is higher than that in control group(t=2.091?3.048?3.424, P<0.05); 3.In the 21-30 and 31-40 years sub-groups, the serum level of visfatin in pilot were significantly higher than control group(t=3.013.1.972,2.000, P<0.05), and in the ?41 years sub-groups, the pilots were showed no significant difference in serum visfatin with the control group(all P<0.05); 4.The pilots among all age groups showed no significant difference in serum RBP4 and visfatin(all P>0.05); 5.For the RBP4 and visfatin change by flying hours.among the groups was not statistically significant(all P>0.05); 6.The frequency distribution of CC?CT?TT genetype in visfatin -1535C> T polymorphism in CHD group were 20.5%?50.0%?29.5%,in pilot group were 25.6%?50.5%?23.9%,in control group were 29.7%?43.1%?26.7%. CHD group and control group, pilot group and control group visfatin-1535C> T polymorphism genotype and allele frequency distribution of the difference was not statistically significant (all P>0.05).Conclusion:The elevated levels of serum RBP4 and visfatin may be a new risk factor for coronary heart disease; Pilots flying factors may lead to elevated serum levels of RBP4 and visfatin.
