Dodartia orientalis Linn is a kind of perennial herbaceous plant,which is also known as Duode grass,Niuhanshui,Zihuaying and so on.Dodartia orientalis Linn is the only species of Dodartia which belonging to Scrophulariaceae family.This plant mainly distributed in Xinjiang,Neimenggu,Sichuang and Gansu Province of China,and other countries including Kazakhstan,Russia,Uzbekistan and so on.Its whole plant has been used in traditional Chinese medicine for the treatment of bronchitis,allergic rhinitis and urticaria.Presently,only a few studies on the chemical constituents of Dodartia orientalis L.have been reported.In this thesis,we investigated the chemical constitutes of the whole herb of Dodartia orientalis Linn.From the 70%EtOH extract,18 compounds were gained by the means of system solvent extraction,silica gel column chromatography,polyamide column chromatography,Sephadex LH-20 column chromatography,macroporous resin column chromatography,preparative thin layer chromatography(PTLC),preparative high performance liquid chromatography(PHPLC)and recrystallization.And 17 compounds' structures were elucidated from spectroscopic data and physico-chemical properties.Those compounds were betulinic acid(1),?-sitosterol(2),2-(4-hydroxyphenyl)ethanol 32-alkyl esters(3),tetracosanoic acid(4),octad ecane acid decanoic ester(5),ursolic acid(6),triacontanol(7),rubiarbonol A(8),triacontane(9),Pomolic acid(10),luteolin(11),tricin(12),chrysoeriol(13),tricin-7-O-(?-glucuronide-6"-butyl ester)(14),selagin(15),specioside(16)and apigenin(17).Among them,tricin-7-O-(?-glucuronide-6"-butyl ester)was obtained as a new compound,and compound 3,4,5,7,9,11,12,13,14,15,17 were first isolated from this species and Dodartia.We also evaluated their anti-tumor activity against two human tumor cell lines HepG2 and HeLa by the means of MTT assay.Compound 1,2,3,6,8,12 and 17 exhibited potent cytotoxicity against human tumor cell lines HepG2 and HeLa.
