Thlaspi arvense Linn, also called "kugai"vegetable,"e-lan" vegetable, is a cruciferous plant in the family of dicotyledons, and it is widely used in the southeast border gansu province. Thlaspi arvense Linn seeds and grass all contain black mustard glucoside, which is decocted in water can benefite vital energy and improve eyesight, and has active effects on liver cleraing,lumbago, etc.This paper using95%ethanol heat and reflux extraction, solution is concentrated to extract, acidified with1%dilute hydrochloric acid, still standing48h, vacuum suction filter removal of impurity, petroleum ether extraction to colorless, adopting different pH and different polarity solvent extraction methods for further separate Thlaspi arvense Linn crude extract, respectively.1. Thlaspi Arvense Linn Extracting Solution of Different pH ValuesTaking adequate Thlaspi arvense Linn crude extracting solution, distribution droplets stronger ammonia water, pH indicator is used to detect at the same time. Until the Thlaspi Arvense Extracting Solution pH3, using chloroform extraction to colorless, and then distribution droplets stronger ammonia water until the Thlaspi Arvense Extracting Solution pH4, using chloroform extraction to colorless, and by this analogy, until the Thlaspi Arvense Extracting Solution pH9, received seven parts, and separated the parts of pH3by column chromatography on silica gel,and elution agent with petroleum ether:ethyl acetate (1:1)(flow of1-2), ethyl acetate:petroleum ether (4:1)(flow of3-4), ethyl acetate (flow of5), ethyl acetate:methanol (4:1)(flow of6-7), ethyl acetate:methanol (3:2)(flow of8) and methanol (flow of9-12) six gradient elution, and then by using High Performance Liquid Chromatography (HPLC) analysis of each flows and Semi-preparative Reversed Phase Liquid Chromatography (EPLC) separation flow of3,4and6, obtained four compounds,(+)-dehydrovomifoliol(1), vomifoliol(2), isoscopoletin(3) and spearmint wood ester B(4), respectively. These four compounds are isolated from this plant for the first time.2. Thlaspi Arvense Linn Extracting Solution Extracted by Different Polar SolventTaking adequate Thlaspi arvense Linn crude extracting solution, distribution droplets stronger ammonia water.Until the Thlaspi Arvense Extracting Solution pH9~10, using petroleum ether ethyl acetate, dichloromethane and n-butyl alcohol extract,respectively, received four parts, and separated the parts of ethyl acetate by column chromatography on silica gel,and elution agent with chloroform (flow of1-3), chloroform:methanol (20:1)(flow of4-22), chloroform:methanol (10:1)(flow of23-32), chloroform: methanol (4:1)(flow of33-40), chloroform:methanol (3:2)(flow of41-45) and methanol (flow of46-48) six gradient elution, and then by using High Performance Liquid Chromatography (HPLC) analysis of each flows and Semi-preparative Reversed Phase Liquid Chromatography (EPLC) separation flow of8,9,12,18and28, obtained six compounds,nobiletin(5), grasshopper ketone(6), dihydrodehydrodiconiferyl alcohol(7), apigenin(8), luteolin(9), blumenol C glucoside(10) and apigenin-7-O-β--D-glucoside(11), respectively. In addition this part also obtained (+)-dehydrovomifoliol(1) and vomifoliol(2). Compounds5~7and10~11are isolated from this plant for the first time.In this paper, a total of16compounds were isolated, the structures of11compounds have been identified. There are three flavonoid compounds: nobiletin(5), luteolin(8) and apigenin(9); Sesquiterpenoids have four:(+)-dehydrovomifoliol(1), vomifoliol (2), grasshopper ketone(6) and blumeol C glucoside(10); Lignans compounds have two:spearmint wood ester B(4) and dihydrodehydrodiconiferyl alcohol(7); Flavonoid glycoside has one: apigenin-7-O-β-D-glucopyranoside(11); Coumarin compounds has one: isoscopoletin(3).The results of antitumor activity in vitro demonstrate:The pH fractions have certain proliferation inhibition for HepG2, R-HepG2and Hep3B, respectively. Chemical compounds of nobiletin(5), luteolin(8) and apigenin(9) have strong proliferation inhibition for HepG2, R-HepG2and Hep3B at the concentration of50μM. The results of bacteriostat experiment demonstrate:The pH3fraction has certain bacteriostatic activity for escherichia coli and bacillus subtilis, The ethyl acetate fraction and dichloromethane fraction have good bacteriostatic activities for escherichia coli.
