Effect Of Tert-Butyl Hydroquinone On The Expression Of Vascular Endothelial Growth Factor And B-Cellleukemia/Lymphoma-2 In Retina Of Type 2 Diabetic Rats

objective: To observe the effect of tert-Butyl Hydroquinone(t B HQ) on vascular endothelial growth factor(VEGF) and the B-cellleukemi a/lymphoma-2(Bcl-2) in retina of type 2 diabetic rats and discuss the po ssible mechanism via Nrf2/ARE signal path. Methods: 50 clean healthy male SD rats were randomly divided into normal control group and model group. The model group were fed with diet high in fat, yolk and sugar for 4 weeks. After 12 hours of fasting, type 2 diabetic rat model was in duced by intraperitoneal injection of low dose of streptozotoin accroding to weight(30mg/kg), then successful modeling rats were randomly divided into diabetic group and tBHQ group. tBHQ was added into the high fat and high sugar diets after the type 2 diabetic model was established for one week in the tBHQ group. The diabetic group were continued to fed with high fat and high suger diet. Respectively, 4 and 12 week after tBHQ treatment, fasting plasma glucose(FPG), total cholesterol(TC), total triglyceride(TG), high-density lipoprotein(HDL-c), low density lipoprotein(L DL-c), fasting serum insulin(FINs) were evaluated. Immunohistochemical method and Real-Time qRT-PCR were used for detecting the content of VEGF and Bcl-2 in retinal of type 2 diabetic rats. Results: Type 2 dia betic model was successfully established in 35 rats, the success rate was 87.5%. The differences are significant of FPG in rats between groups(FGroup=78.531,P=0.000). Comparing with normal group, the FINs of dia betic group and tBHQ group was higher(FGroup=22.480,P=0.000;FTime=7.636,P=0.008). TC, TG of diabetic group was higher than that of normal group and tBHQ group and they were signficently different(TC:FGroup=28.049,P=0.000;TG:FTime=13.108,P=0.000). Comparing with normal g roup, the LDL of diabetic group and tBHQ group was higher(FGroup=6.804,P<0.05). Immunohistochemical showed that VEGF and Bcl-2 were mai nly expressed in retional ganglion cell layer, inner plexiform layer, outer plexiform layer. The expression of VEGF in diabetic group was significa ntly higher than normal group and tBHQ group(FGroup= 11.805,P=0.000). Bcl-2 protein showed an increasing trend in normal group, diabetic grou p, tBHQ group and it expressed more at 12 week than 4 week. The diff erence was significant(FGroup=22.943,P=0.000;FTime=7.170,P=0.08). Bcl-2 protein expressed more in tBHQ group at 12 w than 4w(t=-3.013,P<0.05). Real-Time qRT-PCR showed the m RNA level of VEGF in retinal was significantly higher in diabetic group than normal group and tBHQgroup, the difference of different group was significant(FGroup=79.220,P=0.000;FTime=6.090,P<0.05). The expression of Bcl-2 m RNA in tBHQ group was significantly higher than normal group and diabetic group(FGroup=105.000,P=0.000;FTime=13.17,P<0.05). Besides, tBHQ group at 12 w ex pressed more than the group at 4w(t=-7.047,P<0.05). Conclusion: High sugar, high fat diet combined with small dose of STZ intraperitoneal injection in SD rats can successfully synthesized the type 2 diabetic rats an d diabetic retinopathy rats. tBHQ may have effect of resistance to high blood sugar, adjusting insulin and regulating blood lipid. tBHQ may induce Bcl-2 expression, decrease VEGF expression and thus reduce oxidative injury, increase antiapoptotic effect in retina of diabetic rats.
