ObjectiveTo investigate the protective effect and the mechanism of Fuyuan capsule on Osteoarthritis'apoptosis of cartilage cells and cartilage matrix degradation.MethodsHuman chondrosarcoma cells were cultured and divided into 5 groups randomly:1) normal group,2) model group,3) inhibitor group,4) Fuyuan capsule group,5) Fuyuan capsule+inhibitor group. The percentage of cell cycle and apoptosis index was detected by Flow cytometry. The activity of cell proliferation was detected by CCK-8. The expression of MMP-3,-13 mRNA was determined by Q-PCR. The expression of cell signaling factor of p-JNK and Bax was determined by Western-blot.ResultsThe model group compared with normal group, the percentage of Go/G1 phase increased, the percentage of S phase and G2/M phase decreased, the PI decreased; the apoptosis rate was significantly increased; the activity of cell proliferation decreased; the expression of MMP-3,-13 mRNA andp-JNK, Bax protein were significantly increased (P<0.01). The intervention group compared with the model group, the PI increased; the apoptosis rate increased significantly; the activity of cell proliferation increased; the expression of MMP-3,-13 mRNA and p-JNK, Bax protein were decreased (P<0.01 or P<0.05); the Fuyuan capsule+inhibitor group was most significantly compared with the model group(P<0.01).ConclusionFuyuan capsule can inhibit the apoptosis and promote the proliferation in OA cartilage cells; Fuyuan capsule can inhibit the degradation of extracellular matrix; Fuyuan capsule is stronger than the specific inhibitor of JNK single in the prevention and treatment of osteoarthritis; Its mechanism may be related to the inhibition of the JNK signaling pathway.
