Objective:This research is based on the good retinal protection effect of BuJingYiShi tablet, discusses the Oxidative stress injury and interrelated protective mechanism of radiation retinopathy animal model effected by BuJingYiShi tablet, with building radiation retinopathy model of mice, provides new ideas and methods of prevention and control of radiation retinopathy and similar diseases with TCM.Methods:With using linear accelemiceor to radiate mice eyes, explore radiation retinopathy model of mice which build radiation induction and the best radiological dose. Except the 15 mice in normal control group, the 75 mice would be randomized grouping:model group, dexamethasone group, BuJingYiShi tablet groups with high, medium and low dose and radiated to make radiation retinopathy model of mice.3 months after radiated, with continuous treatment for 1 month, use HE staining to observe the mice retinal morphology; use chemical colorimetric method to measure SOD、MDA content of retina, use ELISA to measure ROS content, use Immunohistochemistry staining method to mesure the VEGF average optical density of retina, use Western blot to measure p53 protein expression of retina.Results:①3 months after modeled, HE staining observation shows that 30Gy group compared with the control, layer structure of retina is loose, arranged in disorder, nuclear swelling and decrease of external granular layer. Compared with normal control group,30Gy group’s vigor of SOD was significantly dropping, the difference was statistically significance (P<0.01); the contant of MDA was significantly rising, the difference was statistically significance (P<0.01); the contant of ROS was significantly rising, the difference was statistically significance (P<0.01); the average potical density in mice’s retina of VEGF was dropping, the difference was statistically significance (P<0.01). This shows that Oxidative stress injury after radiation, Dosages finding study points out that 30Gy is the best radiation modelling dose.② 1 month after treatment, (1) HE staining observation shows that the thickness of the outer nuclear layer of drug group increased. (2) Test the vigor of SOD:Compared with normal control group, the model group、dexamethasone group.. BuJingYiShi tablet groups with high, medium and low dose’s vigor of SOD were lower, the difference was statistically significance (P<0.01-0.05). Compared with the model group, dexamethasone group and BuJingYiShi tablet high does group’s vigor of SOD was higher, the difference was statistically significance(P<0.01-0.05). (3) Test the contant of MDA:Compared with normal control group, the model group、 dexamethasone group、BuJingYiShi tablet groups with high, medium and low dose’s contant of MDA were rising, the difference was statistically significance (P<0.01-0.05). Compared with the model group, the dexamethasone group’s contant of MDA were rising, the difference was statistically significance (P<0.05), BuJingYiShi tablet groups with high, medium and low dose’s contant of MDA were not significantly change, the difference was not statistically significance (P>0.05). (4) Test the contant of ROS:Compared with normal control group, the model group、 dexamethasone group、BuJingYiShi tablet groups with high, medium and low dose’s contant of ROS was significantly rising, the difference was statistically significance (P<0.01). Compared with the model group, dexamethasone group and BuJingYiShi high does group’s contant of ROS were significantly dropping, the difference was statistically significance (P<0.01). Compared with the dexamethasone group, the BuJingYiShi tablet groups with medium and low dose’s contant of ROS were not significantly change, the difference was not statistically significance (P>0.05). This implies that the BuJingYiShi tablet can inhibit the oxidative stress jnjury. (5) Test the average optical density of VEGF:Compared with normal control group, the model group、dexamethasone group、BuJingYiShi tablet groups with high, medium and low dose’s average optical density of VEGF were significantly rising, the difference was statistically significance (P<0.01). Compared with the model group, dexamethasone group and BuJingYiShi high does group’s average optical density of VEGF were dropping, The difference was statistically significance (P<0.05). This implies that the BuJingYiShi Tablet can restrain VEGF expression. (6) Test the average optical density of ICAM-1:Compared with normal control group, the model group、dexamethasone group、BuJingYiShi tablet groups with high, medium and low dose’s average optical density of ICAM-1 were significantly rising, the difference was statistically significance (P<0.01). Compared with the model group, dexamethasone group、 BuJingYiShi tablet groups with high, medium and low dose’s average optical density of ICAM-1 were not significantly change, the difference was not statistically significance(P>0.05). (7) Test the expression of p53 protein:Compared with normal control group, the model group、dexamethasone group、BuJingYiShi tablet groups with high, medium and low dose’s expression of p53 protein were rising, the difference was statistically significance (P<0.01-0.05). Compared with the model group, dexamethasone group and BuJingYiShi tablet high does group’s expression of p53 protein were dropping, the difference was statistically significance (P<0.01-0.05), This implies that the BuJingYiShi tablet can restrain p53 to inhibit the oxidative stress jnjury and VEGF expression.Conclusion:BuJingYiShi tablet tablet can partly restrain early retinopathy of oxidative stress injury、VEGF expression, improve the recovery of oxidative stress injury and reduce the damage of retinal cells. The mechanism of action is related to restraining mice’s retina p53 protein expression, increasing activity of retina SOD and restraining Oxidative stress injury by reducing ROS content, reducing retina VEGF expression.
