Objective: To observe effect of BuJingYiShi tablet on the changes of myelin sheath of optic nerve and expression of MDM2 and p53 in PI3K/Akt pathway of optic nerve in SD Rat Model of Chronic Elevated Intraocular Pressure,and to explore its mechanism.Methods: Thirty SD rats were randomly divided into three groups: control group,model group and treatment group.10 rats in each group.Establishment of chronic EIOP model in rats by cauterization of superior scleral vein.Continuous gastric perfusion for 8 weeks,and all rats were killed at the end of the 8th week.To observe the effect of BuJingYiShi tablet on the myelin sheath of optic nerve and expression of MDM2 and p53 in PI3K/Akt pathway of optic nerve in Rat Model of Chronic Elevated Intraocular Pressure.Results:(1)IOP:Compared the IOP of rats in model group and treatment group immediately and 8 weeks after cauterization of superior scleral vein with before cauterization,the IOP was significantly higher,and the difference was statistically significant(P<0.05).The IOP in the model group immediately after cauterization was comparable to that in the 8th week after cauterization,there was no statistically significant difference(P>0.05).The IOP in the treatment group was higher immediately after cauterization than 8 weeks after cauterization,and the difference was statistically significant(P<0.05).(2)Changes of myelin sheath of optic nerve as follows:The myelin sheath of optic nerve of total area and integrated optical density in treatment group(3.0091±0.216,83776.59±17932.626?m2)compared with control group(3.4574±0.0870?m2,109844.86±19399.470?m2),it was obviously low,and the difference was statistically significant(all P<0.05);Compared with model group(2.4988±0.186?m2,38018.80±11826.286?m2),it was obviously high,and the difference was statistically significant(all P<0.05).(3)MDM2 expression in optic nerve as follows:Total area and integrated optical density in treatment group(0.1625±0.026?m2,2086.70±744.326?m2)compared with control group(0.2604±0.047?m2,3548.57±906.465?m2),it was obviously low,and the difference was statistically significant(all P<0.05);Compared with model group(0.0910±0.027?m2,1195.86±334.984?m2)it,was obviously high,and the difference was statistically significant(all P<0.05).(4)p53 expression in optic nerve as follows: Total area and integrated optical density in treatment group(2.6553±0.396?m2,96419.47±38603.142?m2)compared with control group(1.9827±0.350?m2,29393.33±9977.039?m2),it was obviously high,and the difference was statistically significant(all P<0.05);Compared with model group(3.3768±0.168?m2,345824.79±56678.048?m2),it was obviously low,and the difference was statistically significant(all P<0.05).Conclusion:The rat model of chronic EIOP established by cauterization of closed scleral vein is easy to operate,and the effect of increasing intraocular pressure is stable.High intraocular pressure can be obtained more than 8 weeks.Bujinyishi tablet can effectively protect the optic nerve and visual function of chronic EIOP rat model.Its function is as follows:Slight reduction of intraocular pressure;Increase the content of myelin sheath of optic nerve;and it may up-regulate the expression of MDM2 in PI3K/Akt pathway of optic nerve and down-regulate the expression of p53 in PI3K/Akt pathway of optic nerve.
