This paper is focus on extraction technology of Dioscorea nipponica Makino,and alcohol precipitation of concentrated solution. The subject comes from Tianjin Shijitianlong Pharmaceutical Co. Ltd. Longjiatongluo capsule is one of its products. Dioscorea nipponica Makino is one of the main materials of Longjiatongluo capsule, and the extraction of Dioscorea nipponica Makino is a traditional method. Based on different experimental design and evaluate indexes, we used different optimization method to optimize the process of extraction and alcohol precipitation, respectively. Through the experiment, we select the best technology of extraction and alcohol precipitation. We also tried the scale-up produce preliminary. The main work was as following:In this paper, we tried to optimize the water extraction process of rhizomes of Dioscorea nipponica Makino based on dynamic reflux extraction. An experiment in accordance with a central composite design was carried out on the water extraction. The aqueous extraction contained a large amount of non-active constituents, so an alcohol precipitation(AP) was applied. The optimal parameters obtained by response surface methodology(RSM) were as follows: solvent: solid: 5:1, speed of back-flow: 6ml/min, back-flow time: 3.94 h. Under these optimum conditions, the yield of Dioscin was 7.576 mg from 1g of Dioscorea nipponica Makino,the dry extract was 0.2145g/g. In the same with water extraction, we also did a research about alcohol extraction process of rhizomes of Dioscorea nipponica Makino based on dynamic reflux extraction. The technology is solvent/solid: 7.6 m L/g, back-flow time: 4.8h, speed of back-flow: 10.3 m L/min, the yield of Dioscin is 13.83mg/g, the dry extract is 0.1849g/g. In lab, we also use the transfer rate of Dioscin as the evaluation index to compare the technology from the factory?the technology in lab?and the alcohol extract. The result is 19.76%? 33.26%? 83.31%.Alcohol precipitation is a traditional method to purify the water extraction. The research on alcohol precipitation of water extraction of Dioscorea nipponica Makino has been carried out. The optimum of technology is relative density=1.08, ethanol concentration = 70%, temperature of alcohol precipitation=20?, temperature of standing=20?,the rate of dioscin transfer= 83.77%, the rate of removing impurity= 34.67%...
