The Research Of Casein Glycomacropeptide(CGMP) On The Signaling Pathways Of Intestinal Mucosal Acquired Immunity

Casein Glycomacropeptide(CGMP)is a polypeptide from ?-casein in milk,CGMP has many bioactive functions that have been proved by many researchers.This study was based on the preceding knowledge provided by other men,which was aimed to study the immunoregulation effects on the innate immunity and acquired immunity of CGMP by stimulating with CGMP.The research was divided into two parts:on the one hand,using Balb/c mouse as laboratory animals,treating them with different concentrations pure CGMP by oral administration or intraperitoneal injection,after two hours,sampling blood from the eye sockets of the mouse.Then separating serum from blood and numbering them.Using Millipore mouse Cytokine kits to test the concentrations of 24 cytokines in serum of mouse.The concentrations of CGMP were used to study the effects of CGMP on the cytokine network and the impacts of innate immune responses.And the results indicated that the remarkable change of cytokines after treating with CGMP by oral were: GM-CSF?IFN-??IL-2?MIP-1?;while the cytokines were Eotaxin?TNF-??G-CSF?GM-CSF?IL-6?IL-17?KC treating with CGMP by intraperitoneal injection.On the other hand,dendritic cells derived from bone marrow,which was separated from male C57BL/6 mouse,was stimulated with different concentrations of CGMP under cultivation after sometimes in vitro,and complete medium and LPS was regarded as blank control and positive control separately.When cultivation finished,testing the maturation of DCs on Flow Cytometry;the stimulated DCs was cultured with T cells in different ratio for 96 hours,then testing the proliferation of T cells with CCK-8 kit.Using the best ratio of DCs and T cells,doing the Mixed Lymphocytes Reaction(MLR)again with the DCs stimulated with different concentrations of CGMP and T cells.After MLR,test the 24 kinds of cytokines in the culture supernatant.The results showed that each concentration of CGMP could stimulate the maturation of DCs while lower than the positive control.The results of MLR indicated that T cells were proliferated by culturing with DCs.And the remarkable change of cytokines in the culture supernatant were G-CSF?GM-CSF?IFN-??IL-1??IL-2?IL-4?IL-5?IP-10.On the whole,the study indicated that DCs could present some peptides to T cells which leaded related immunocytes activation and proliferation.
