1 ObjectiveThis research topics is based on the "kidney essence-brain", "blood-blood stasis" and other related theory. Using the Wnt signaling regulates research as a breakthrough point, Mainly through Naoluoxintong recipe and Bushenshengsui recipe, making MCAO/R rats as an animal model for experimental studies.The rats intervention includes the Cerebral ischemia and reperfusion. Protein expression of GSK3?, Sfrp1 and Wnt9a is observed in hippocampus and frontoparietal cortex of model rats. From the view point of molecular biology, yiqi huoxue method and kidney raw pulp method is explained on the basis of experimental results and the related experimental data.2 MethodFirst, choosing 56 healthy Sprague-Dawley(SD) rats of male, four months,average weight between 280 g and 320 g. This 56 rats are randomly divided into four groups: sham group, model group, Yiqihuoxue method group, Bushenshengsui method group, each group has 14 rats. Making each group of rats, then each group of rats are randomly divided into two groups for 3d and 7d group, each group has 7 rats.Establishing the right of focal cerebral ischemia rat model by modified suture method,took Naoluoxintong recipe and Bushenshengsui recipe moderate dose: 8.54g/kg,12.87g/kg. The rats of Yiqihuoxue method group and Bushenshengsui method group are administrated for half an hour before making MCAO/R rat model. After this step, the rats are administrated between 9:00 and 16:00 everyday, continuous administrated of 3d,7d. Sham group and model group are given the same dose of normal saline. Selecting fresh ischemic hippocampus and frontoparietal cortex side of rats, the protein expression changes of GSK3?, Sfrp1 and Wnt9 a in ischemic hippocampus and frontoparietal cortex are analyzed by Western-Blot. After color film scanning, thesepictures are analyzed by professional software Image J. The data was analyzed by SPSS20.0. The analysis results is drew by VISIO 2007.3 ResultWestern Blot results showed that: GSK3?strip is located at the 46 / 51 kda place,Sfrp1 strip is located at the 33 kda place, Wnt9 a strip is located at the 37 kda place,Actin strip is located at the 42 / 44 kda place.The effects of 3d Yiqi Huoxue method group and 3d Bushen Shengsui method group on the protein expression of GSK3?, Sfrp1 and Wnt9 a in ischemic hippocampus and frontoparietal cortex: Compared with sham group, the model group protein expression of three proteins are obviously increased(P<0.01); Compared with model group, the Yiqi Huoxue method group and Bushen Shengsui method group protein expression of three proteins are obviously reduced(P<0.01); Compared with Bushen Shengsui method group, the Yiqi Huoxue method group protein expression of three proteins are obviously reduced(P<0.01). The result of the frontoparietal cortex is consistent with the hippocampus.The effects of 7d Yiqi Huoxue method group and 7d Bushen Shengsui method group on the protein expression of GSK3?, Sfrp1 and Wnt9 a in ischemic hippocampus and frontoparietal cortex: Compared with sham group, the model group protein expression of three proteins are obviously increased(P<0.01); Compared with model group, the Yiqi Huoxue method group and Bushen Shengsui method group protein expression of three proteins are obviously reduced(P<0.01); There is no significant difference between Yiqi Huoxue method group and Bushen Shengsui method group(P>0.05). The result of the frontoparietal cortex is consistent with the hippocampus.4 ConclusionYiqi Huoxue formula and Bushen Shengsui formula can promote the repairment of injured cerebral tissue by the way of reducing the protein expression of GSK3 a,Sfrp1and Wnt9 a in hippocampus to regulate Wnt/?-catenin signal pathway.
