Compound formula is the main form of Traditional Chinese Herbal and the core of the compound is the compatibility between different drugs.The clinical application of "Toxic medicine" was often certainly restricted because of its side effects on bodies, but never be given upfor the clinical efficacy and irreplaceable for people dismay.Tripterygium wilfordii was applied for the refractory disease treatment, such as rheumatoid arthritis, chronic nephritis, and widely used in the clinical. However, a series of toxic and side effect was emerging with the increased use and deeply study,hepatotoxicity, renal toxicity, and so on. Xinfeng capsule, a hospital traditional Chinese herbal formula of The First Affiliated Hospital of Anhui University of Chinese Medicine(Anhui medicine system Z20050062) consists of four herbs, which are astragalus,myotonin, thunder god vine and centipede. Clinical studies have identified that Xinfeng capsule has unique advantages and less side effects.On the basis of preliminary pharmacological experiments, concentrations of triptolide and celastrol in Tripterygium wilfordii combination groups were determined by HPLC binding component-knockout approach. The main ingredients were analyzed to find the difference between single herbs and the best combination group,which aimed at filtrating the best compatibility group to decrease the toxic, and understanding the new method and idea to decrease the toxicity by investigating the key component change before and after drug combination, and increasing the clinical safety.Objective:1. To establish the measure method of triptolide and celastrol in Tripterygium wilfordii.2. To establish the measure method of triptolide and celastrol in Xinfeng capsule.Study on the regularity of triptolide and celastrol dissolution in Xinfeng Capsule decoction to find the chemical basis related to Tripterygium wilfordii compatibility for attenuation.3. To establish the fingerprint of the best combination groupand find the difference between single herbs and the combination group.Methods:1. Consulting literature material, screened the best extraction method of Tripterygiumwilfordii.2. Study chromatogram conditions, mobile phase, detection wavelength, establish the measure method of triptolide and celastrol using HPLC-DAD method.3. To establish the fingerprint of TR-group(Tripterygium wilfordii and Radix Astragal group) with T-group(Tripterygium wilfordii group). The main ingredients were analyzed to find the difference between single herbs and the best combination group to decrease the toxic of Tripterygium wilfordii.Results:1. According to the document retrieval results, summarized the optimal extraction method of Tripterygium wilfordii was methanol extraction, then purification using silica gel column and using chloroform- methanol(96:4)to elution.2. All separation steps were carried out on an Agilent Eclipse plus C18 column(4.6mm×250 mm, 5 ?m). The mobile phase of triptolidedetermination was composed of water(A) and acetonitrile(B) using a ratio of 34?66. The flow rate of the mobile phase was kept at 1 ml/min, the column temperature was kept at 30?, the wavelength was set at 220 nmand the sample injection volume was 10 ?l.The mobile phase of celastroldetermination was composed of 1% phosphoricacid aqueous solution(A) and acetonitrile(B)(15:85). The flow rate of the mobile phase was kept at 1 ml/min, the column temperature was kept at 30?, the wavelength was set at 220 nmand the sample injection volume was 10 ?l.The statistics analysis showed that Radix Astragali significantly reduced the dissolving action of triptolide and celastrol, myotonin and centipede compatibility had no significant effect on the dissolving action of them.3. Comparing TR-group(Tripterygium wilfordii and Radix Astragal group) with T-group(Tripterygium wilfordii group) and R-group(Radix Astragal group), the areas of 9 peaks remain the same, while the areas of 3 peaks were increased and 18 peaks were decreased, there were four new peaks produced.Conclusion:Based on the dissolution changes of chemical components of Tripterygium wilfordii due to compatibility in Xinfeng Capsule, it could be preliminarily inferred that Tripterygium wilfordii induced toxicity could be decreased by combination of Tripterygium wilfordii and Radix Astragal, and compatibility between the drug ingredients are not only a simple addition, but also include qualitative and quantitative changes.
