Objective: Through the observation of the experimental study of recurrent genital herpes in mice and from Longdan Xiegan granules on recurrent genital herpes in mice to evaluate the efficacy of; of Longdan Xiegan granules of recurrent genital herpes in mice peripheral blood dendritic cells(DCS) in tlr1 to TLR10 m RNA expression of the influence. To determine the role of anti HSV virus, and systematically study the mechanism of the action of the Toll receptor signal transduction pathway on the surface of the cell surface. According to the mechanism of antiviral action, from the point of view of immunology, a clear target is put forward to further clarify the mechanism and molecular basis of traditional Chinese medicine in stimulating the innate and acquired immune responses.Methods: Selected 120 female mice were recurrent genital herpes modeling and randomized for six groups, respectively, for traditional Chinese medicine of Longdan Xiegan granule low-dose treatment group, Longdan Xiegan granules dose treatment groups,Longdan Xiegan granules high dose treatment group, aciclovir in irradiation group and normal saline control group and normal control group. To observe the changes of local skin lesions in mice before and after administration, and to determine the curative effect. And on the second day before administration of 1 day of treatment and after the end of, the extraction of mice in each group of jugular vein blood 2ml, with lymphocyte separation liquid separation of peripheral blood mononuclear cells in vitro by GM-CSF, IL-4, TNF alpha induced to generate a large number of mature dendritic cells, by fluorescence quantitative RT-PCR method to detect mice before and after the treatment of peripheral blood dendritic cells(DCS) in tlr1 to TLR10 m RNA expression.Results: 1. Comparing with normal control group compared to the model group before treatment visible lesions and pathological state of obvious, the difference is statistically significant(P < 0.05); lesion score before treatment between the model group had no significant difference(P > 0.05). After treatment, the differences between the traditional Chinese medicine Western medicine group and high dose group have statistical significance(P < 0.05), western medicine group and Chinese medicine group of low concentration difference between no statistical significance(P > 0.05); after treatment between the traditional Chinese medicine in the treatment group, the difference has statistical significance(P < 0.05) and traditional Chinese medicine in the treatment group with different treatment, Chinese medicine concentration group effect best, high concentration times. Effect of low concentration group is the worst, the effect of low concentrations of ACV and traditional Chinese medicine group similar. Longdan Xiegan Decoction group, the concentrations of total efficiency is 95%, low concentration group and acyclovir group, the total efficiency of the same, between groups of efficiency are different, with statistical significance(P < 0.01). 2. The model group before treatment TLRm RNA level compared with normal control group, the difference had statistical significance(P < 0.05), model group compared with normal control group, tlr1, TLR4, TLR6, TLR7, tlr8, TLR9 and TLR10 level decreased, increased TLR2, TLR3 and TLR5, My D88, NF KB level; the model of dendritic cells in group of CD11 c, CD80, CD83, CD86 and HLA-DR, HLA-ABC was significantly higher than that of control group(P < 0.05). After the treatment of peripheral blood dendritic cells TLRm RNA expression level in the normal control group and the treatment group had significant difference(P < 0.05), the treatment group tlr1, TLR4, TLR6, TLR7, tlr8, TLR9 and TLR10 level increased, decreased TLR2, TLR3 and TLR5, My D88, NF KB level; after treatment of dendritic cell surface molecule CD11 c, CD80, CD83, CD86 and HLA-DR, HLA-ABC trend to normal(P > 0.05).Conclusion: Longdan Xiegan granules for complex lesions in mice of hair genital bleb has exact curative effect, Longdan Xiegan granules could achieve in the immune signal transduction in cells through the regulation of peripheral blood dendritic cells TLRm RNA level, the release of synthesis of the corresponding cytokines, induction of dendritic cell differentiation and proliferation, the start cell immunity against the virus.
