Objective:By establishing severe acute pancreatitis(SAP)model in Sprague-Dawley rats,we observe the pathology changes of pancreas and intestinal and the serum of inflammatory mediators TNF-?,IL-6.In our study,compares the change of small intestine tissue homogenate TNF-?,IL-6 and intestinal mucous SIgA.Expect to discuss the change of inflammatory factors in SAP and explore the effection ofinflammatory mediators and intestinal mucosal barrier damage in SAP.Methods:Fourty-eight healthy adult male SD rats,weight 260-300 g were randomly divdied into two small groups,sham-operation(A group N=24),SAP(B group N=24).Each group as the phases of 6h,12 h,24h.Each small group was eight rats.Before experiment,Each rat fasting 12 h free drinking water.SAP group:SAP model was established in rats by injecting 5% sodium taurocholate into biliopancreratic duct.A group:After laparotomy in physiological saline(0.1 ml/100 g weight)it is inject.To supplement the fluid loss in rats leg subcutaneous injection of 0.9% sodium chloride solution(2ml/kg)postoperative still fasting rats,free drinking water after awake.Each group 24 rats as the phases of 6h,12 h,24h,each point in the time only 8 rats been caesarean section.Macroscopic observation of pancreas and intestinal largely changed,After abdominal aorta collected blood samples 5 ml,centrifugal in serum,serum samples preserved in-20?,used for subsequent detection,automatic biochemical analyzer method to detect the serum amylase.Use the ELISA method for determination of serum TNF-?,IL-6 and intestinal tissue homogenate TNF-?,IL-6 and small intestinal mucous SIgA.The pancreas and ileum samples were processed for microscopic histologic lesion score.The experimental data from using SPSS17.0statistical software for statistical analysis.Results:1.SAP group compared to control each point in time the serum levels of AMS,IL-6,TNF-? were significantly increased in group SAP(P<0.01).2.SAP group at each time point the pathological scores of pancreas and ileum tissue injury were markedly increased in group SAP(P<0.01),SAP group of pancreas and ileum histopathological grading increases with time extend.3.SAP group of rat intestinal mucous SIgA at each time point were lower than control group.4.The level of TNF-? and IL-6 in intestinal mucosa and histopathological scores of intenal mucosa were all significantly increaed but SIgA content was decreased in model group at each time point after establishment of model as compared with the sham operated group.5.There was a positive relationship between inflammatory mediators(TNF-?,1L-6)in intestinal mucosa and histopathological scores of intestinal mucosa(P <0.01).There was a negative relationship between inflammatory mediators(TNF-?,IL-6)and SIgA content in intestinal mucus(P < 0.01).Conclusion:1.Rats of SAP were induced by retrograde injection of 5% sodium tatiroeholate into the biliopancreatic duct,this experimental method is stable and repeatable.2.Cytokine TNF-? and IL-6 involved in the occurrence and development of SAP,the level of TNF-? and IL-6 may be as predictors of prognosis of acute pancreatitis.3.SAP rats can happen early intestinal mucosal barrier dysfunction,impaired immune barrier,characterized by SIgA decline.4.Severe acute pancreatitis not only cause pancreatic injury but also lead to the damage of the intestinal mucosal immune barrier.The damage is aggravating with the time.The damage is manifested by reducing the secretion of slg A and increasing the intestinal local inflammation factor TNF-?? IL-6.
