Objective: To determine the authenticity and quality of Zhuang medicine Stenoloma chusanum Ching and establish a preliminary quality valuation for it.Methods: Make clear the history evolution of Stenoloma chusanum Ching by herbal textual research.Pharmacognostic identification studies were conducted,including identification of the original plant,description of the characters and physical and chemical identification.To authenticate Stenoloma chusanum Ching and Onychium japonicum(Thunb.)Kze.with microscopic identification.According to ChP(2015),the content limits of water,ash and alcohol extract were determined.Contents of four ingredients in STENOLOMATIS CHUSANI HERBA(SCH)was determined with quantitative analysis of multi-components by single marker(QAMS)method.Fingerprint of Chinese materia medica(CMM)was used to evaluate its quality.Results:(1)The species and function of Stenoloma chusanum Ching were figured out which provides theoretical basis for clinical application and scientific research.(2)The original plant morphology,medicinal materials characteristics,and thin layer chromatography(TLC)were useful in identifying SCH.(3)The spore of Stenoloma chusanum Ching is ovate-spherical with one side hollow in shape while Onychium japonicum(Thunb.)Kze.'s looks like spherical or subtriangular tetrahedron.Stenoloma chusanum Ching's rhizome has amphicribral siphonostele with pith tissue in the middle.Onychium japonicum(Thunb.)Kze.has amphicribral meristele.The petiole of Stenoloma chusanum Ching has one “U” shaped vascular bundle but Onychium japonicum(Thunb.)Kze.has two without “U” shape.Foliage epidermis cells of the former are thin walls however the latter's are a little thick walls.(4)In the root and rhizome,water content ranges from 6.64% to 10.35%,total ash content is among 8.50% to 15.91%,acid-insoluble ash content is 4.98% to 11.10%,alcohol extract content ranges from 7.95% to 21.40%.In the hole herb,water content ranges from 7.45% to 12.24%,total ash content is among 5.04% to 11.29%,acid-insoluble ash content is 1.39% to 6.58%,alcohol extract content ranges from 24.96% to 38.05%.In the leaves and petiole,water content ranges from 8.35% to 15.62%,total ash content is among 3.94% to 5.00%,acid-insoluble ash content is 0.17% to 0.80%,alcohol extract content ranges from 26.61% to 35.76%.(5)The results of QAMS method were validated and no obvious significant difference by comparing with that of external standard method.The relative correction factors(RCF)of four gradients were good reproducibility.(6)The common mode of HPLC fingerprints for SCH were set up.There were 14 common peaks in the fingerprints and four main peaks were identified.Conclusion: Based on the above method and data,primary quality evaluation of the Zhuang medicine herb Stenoloma chusanum Ching is established,laying a foundation of safety,effectiveness and controllability for its clinical application.There have been no similar domestic and international documents in this field,which has been proved by authority.
