ObjectiveTo select the optimum preparation technology of hyaluronic acid oligosaccharide ointment,to prepare of hyaluronic acid oligosaccharide ointment,to study its quality standard,and to observe the effect of hyaluronic acid oligosaccharide ointment wound healing in order to provide the scientific basis for further clinical trials.MethodsScreening of commonly used cream base vaseline,lanolin,triethanolamine and stearic acid,glycerol monostearate,liquid paraffin,glycerin,sodium dodecyl sulfate,preservatives and so on.By observing the soft paste appearance,malleable,stability to determin preliminary of ointments types and dosage range of accessories.glycerol monostearate,sodium dodecyl sulfate,the amount of liquid paraffin is optimized to get the best prescription by orthogonal design method.Hyaluronic acid oligosaccharide content of oligosaccharides hyaluronic acid ointment was determined by carbazole method.To prepare of rabbit ear skin defect model,full-thickness skin defect was at 3 sites on the ear skin,and coated with 3 different concentrations of hyaluronic acid oligosaccharides ointment,blank matrix,snow cream ointment?positive drug?as well as blank control,respectively.To observe the effect of hyaluronic acid oligosaccharide ointment on New Zealand rabbits and on days 4,7 and 14 after the injury,the healing area and the healing rate for each wound were determined,On day 14 after the injury the tissues around the wounds were sampled for pathological examination and TGF-?,VEGF,IGF protein expression was detected by immunohistochemistry.ResultsWith the orthogonal design,to determine the ointment optimal prescription as: Huang Fanshilin 2.00g,Stearic acid glyceride2.10g,Glycerin monostearate 1.60g,Liquid paraffing 2.50g,Methyl neper gold 0.02g was used as oil phase and glycerin 2.00g,Sodium dodecyl sulfate 0.20g.Distilled water 9.60mL was used as water phase.The main drugs: hyaluronic acid oligosaccharide is about 0.06g.Ointment is delicate and easy to coating,In 55 ? oven and-20 ? refrigerator placed 6h,24h,respectively,it does not appear oil-water stratified phenomenon and centrifuge at the speed of 2500r·min-1 is stable and no oil-water stratified demulsification phenomenon,suggested that it is stable in high temperature or low temperature.p H is not greater than 8.3 of 3 batches of ointment according to the requirements of the pharmacopoeia.Sulfuric acid carbazole method of content determination results is reliable.HE staining and immunohistochemical staining showed that hyaluronic acid oligosaccharide ointment group,positive drug group had statistically significant difference compared to the blank substrate group and suggested they had a significant effect on wound healing.Base grop compared with blank grop had no statistical significance,so ruled out the interference of the cream base,and suggested hyaluronic acid oligosaccharide play the leading role in promoting wound healing process.ConclusionsThe quality of the hyaluronic acid oligosaccharide ointment makes accords with the ointment demands in 2015 edition chinese pharmacopoeia.And this ointment have obvious improvement on the wound healing rate and quality.It is showed that the ointment has healing effect on the wound healing.
