| The dissertation described phytochemical investigation on diterpenoids from Euphorbia helioscopia, and flavonoids from the flowers of Coreopsis tinctoria.Euphorbia helioscopia L. belongs to the genus of Euphorbia of the family Euphorbiaceae, a traditional Chinese medicine(TMC) for the treatment of chronic obstructive pulmonary diseases such as cough, phlegm-turbidity, asthma, and chronic bronchitis. Pharmacological research showed diterpenoids from E. helioscopia had prominent anticancer activities. The pieces of the aerial part of E. helioscopia were extracted with 95% methanol. Twenty-two diterpenoids were isolated from the EtOAc fraction, and were identified to be diverse diterpenoids with ent-abietane, lathyrane, ent-kaurane, ent-atisane and ingenane types, which included seventeen new ones(1-17) and five known ones(18-22), along with twelve other kinds of compounds(23-34), including one new apocarotenoid(23) and one new coumarinolignoid(27), were isolated from the extract.Coreopsis tinctoria Nutt. belongs to the genus of Coreopsis of the family Compositae, the original plant of Chinese Uygur folk medicinal herb ‘Snow chrysanthemum’, which is named ‘Shemuju’ in the Xinhua Herbal Scheme, with the effects of heat-cleaning, detoxicating, dampness-dissipating and dysentery-curing. Modern pharmacological studies have demonstrated that C. tinctoria has hypoglycemic, anti-oxidative, hypotensive, hypolipidemic activities, which indicated potential medicinal value of this material. The flowers of C. tinctoria were extracted with 70% methanol. Eight flavonoids were isolated from the EtOAc fraction, and were identified to be diverse flavonoids with five flavonones(35-39), one flavanonol(40), one chalcone(41) and one flavonol(42), along with four other kinds of compounds(43-46), were isolated from the extract.The structures of the isolated compounds were elucidated by means of integrated analysis of spectral data, including MS(LR- and HR-MS), IR, UV, optical rotations, 1D(1H and 13C), 2D(HSQC, HMBC, COSY and NEOSY) NMR, ECD and X-ray technologies, and comparison with those reported in literatures.The isolated diterpenoids were elvaluated for the cytotoxic effect angainst human breast cancer cell line MCF-7 and human pancreatic cell line PANC-1, while euphelioscoids F(6) and L(12) exhibited moderate cytotoxic activities against MCF-7(IC50 9.5 and 9.8 μM, respectively) and PANC-1(IC50 10.7 and 10.3 μM, respectively) cells. |
