Objective:Through detection of expression of AFP mRNA,h-TERT mRNA,CD44v6 mRNA and MAGE mRNA in patients with primary hepatocellular carcinoma,to study the value of the combination detection of four markers in predicting the early recurrence and metastasis after Hepatectomy?Methods: Peripheral blood samples were obtained from 40 Primary hepatocellular carcinoma and controls,Reverse Transcription-Polymerase Chain Reaction(RT-PCR)was used to detect serum AFP mRNA?h-TERT mRNA?CD44v6 mRNA and MAGE-1 mRNA ?Results:The positive rate of AFP mRNA?h-TERT mRNA?CD44v6 mRNA?MAGE-1 mRNA was 50%(20/40)?62.5%(25/40)?55%(22/40)?37.5%(15/40)?The positive rate for at least one of the four markers was 90%?The combinatorial expression of AFP/hTERT/CD44v6/MAGE-1 has larger area under ROC curve than either AFP mRNA,hTERT mRNA,CD44v6 mRNA or MAGE-1 mRNA(AUC=0.766),and has higher Specificity(96.2%),positive predictive value(88.9%),negative predictive value(71.4%)?In Kaplan-Meier analysis,The clinical stage,hTERT mRNA,CD44v6 mRNA,AFP/hTERT/CD44v6/MAGE-1 mRNA were correlated with postoperative recurrence and metastasis,logistic regression analysis showed that clinical stage and expression of AFP / hTERT / CD44v6 / MAGE-1 were independent predictors of postoperative recurrence in patients with primary liver cancer(P <0.05)?Conclusion: The combinatorial detection of AFP mRNA?h-TERT mRNA?CD44v6 mRNA and MAGE mRNAexpression increased the accuracy of predicting PHC recurrence in patients with early postoperative and maybe has certain clinical application value?...
