Study On Cytoprotective Antioxidant Effects Of Huangqi And Baishao In Synergistic

Compatibility is a common means of traditional Chinese medicine, Which has a very important significance in the use of Traditional Chinese Medcine. Astragalus-White peony is a classic drug pair, its application is very estensive,its efficacy is exact, with a high clinical application value. First,we verify the Astragalus, the White peony different solvent extraction phase's antioxidant capacity using DPPH radical scavenging experiments. Then, we measured the total phenols, flavonoids content and analyzed the relationship between the content of total phenols, flavonoids and antioxidant capacity. Finally,we studied the repairing effect of Astraglas, White peony on cell injury induced by H2O2.Firstly, we measured the antioxidant activity of Astragalus, Penoy root different solvent extraction phases by DPPH radical scavenging method. we found that all extract extractions have DPPH radical scavenging capacity, For Astragalus extract extractions, chloroform phase and ethyl acetate phase have a strong DPPH radical scavenging capacity, However, capacity are between them. For Penoy root extract extractions,ethyl acetate phase has the strongest radical scavenging capacity,water phase has the weakest scavenging activity, the other phases have a midial radical scavenging activity between them. Complex of Astragalus and penoy root different solvent extractions can improve the DPPH radical scavenging capacity, which has a significant difference compared with the theoretical value (P<0.05).This indicates that the complex of different solvent extractions can enhance the antioxidant capacity of each phase in a synergistic way.We also measured the content of total phenols and flavonoids of Astragalus and penoy root solvent extractions.it was found that the content of phenols of Penoy root is higher than Astragalus, while the content of flavonoids is lower. The variation of the coetent in Astragalus and penoy root solvent extractions are the same,as ethyl acetate phase> cloroform phase> n-butanol phase> petroleum ether phase> water phase compounding, the total phenols and flavonoids contents has a significant growth, indicating that the compounding of different solvent extractions increased phase content of phenols and flavonoids in a synergistic way.The MTT assay showed that, in a particular concentration range, Astragalus and Penoy root solvent extractions have repairing effect on cell injury induced by H2O2, when used in compounding, the repairing effect is better. By measuring the activity of SOD and GSH-Px and content of MDA, we found that extract phases can effectively enhance the damaged cell's activities of SOD and GSH-Px, decrease the content of MDA. This proved the positive role of traditional Chinese medicine in response to oxidative stress at the cellular level.
