Depression is a common mental disorder which can cause both mental and physical damages.This disease is mainly characterized by low mood and lack of happiness,and is accompanied by fatigue,lack of attention,sleep disorders as well as cognitive and language dysfunction.Because of its high incidence,recurrent and suicide rate,depression has become one of the public healthy common concern in the world.Currently,clinical studies cannot fully elucidate the pathogenesis of depression and clinical research of depression has many limitations,researches are needed to complete using animal models,so animal models in the studying of the pathogenesis of depression or exploring antidepressant drugs exploring are very necessary.We applied a metabolomics approach based on UHPLC-Q-TOF-MS in chronic unpredictable mild stress model,to investigate e effects of Lilies and Rhizoma Anemarrhenae on a rat experimental model of CUMS and a mice experimental model of global cerebral ischemia and seek the potential biomarkers related to depression induced by the two models.At the same time,we evaluated the therapeutic effect of Anemarrhena Rhizoma,Lilies,and combination of them to further explore possible mechanisms of saponins of Anemarrhena Rhizoma and Lilies.1.We applied a rat model of chronic unpredictable mild,and used the saponins of Anemarrhena Rhizoma and Lilies to intervene in the rat model.Metabonomics method based on UHPLC-Q-TOF-MS was applied to seek biomarkers associated with depression induced by chronic unpredictable mild stimulation.Finally,we screened 31 and 32 differential metabolites respectively from the rats' serum and hippocampus.These metabolites mainly related to glutamine-glutamic acid metabolism,tricarboxylic acid cycle(TCA cycle),phenylalanine,tyrosine,tryptophan metabolism,glycerolphospholipid metabolism,arachidonic acid metabolism,valine,leucine,isoleucine biosynthesis and so on.At the same time,we evaluated effects of Anemarrhena Rhizoma,Lilies saponins based on these differential metabolites and the performance in the behavior tests of rats,combination of saponins of Anemarrhena Rhizoma and Lilies hizoma showed a better effect for the treatment of depression induced by CUMS.2.A mice experimental model of global cerebral ischemia was applied and we used the saponins of Anemarrhena Rhizoma and Lilies to intervene the model.The metabolic profile analysis based on UHPLC-Q-TOF-MS was applied to seek biomarkers related to depression induced by global cerebral ischemia.Finally,we screened 16 and 43 differential metabolites respectively from the mice serum and hippocampus.These metabolites mainly related to Metabolism of phenylalanine,tyrosine,tryptophan,valine,leucine,metabolism,alanine,proline metabolism,taurine metabolism,oxidative stress,etc.Meanwhile,we evaluate effects of Anemarrhena Rhizoma,Lilies saponins based on these differential metabolites and the performance in the behavior tests of mice,the results showed that combination of saponins of Anemarrhena Rhizoma and Lilies had a better effect for the treatment of depression induced by global cerebral ischemia.In conclusion,this study applied metabonomics method to study the antidepresant effects of saponins of Anemarrhena Rhizoma and Lilies on a rat model of chronic unpredictable mild and a mice model of global cerebral ischemia induced depression,the results showed that combination of saponins of Anemarrhena Rhizoma and Lilies has a better therapeutic effect on these two kinds of depression model.This study provided foundation for further clarifying the targets of saponins of Anemarrhena Rhizoma and Lilies.
