The Induction Effect Of Patients Of Hypertension Disease Integrated With QDBS (Qi Deficiency And Blood Stasis Syndrome) Blood Serum On Vascular Endothelial Cells’ Apoptosis And Its Mechanism

Objective: To discuss that the serum of hypertension patients with Qi deficiency and blood stasis(QDBS)whether can arouse apoptosis of endothelial cell or not and analyze related mechanism.At the same time,to observe the intervention effect of Bu yang huan wu soup and verify "formula-syndrome relation".Methods:collecting all kinds of serum and according to the experimental program,these serum is divided into six groups,named respectively hypertension QDBS group,non QDBS group,healthy groups,Bu yang huan wu decoction high,medium and low dose group.All serum above act on human umbilical vein endothelial cells(HUVEC)cultured in vitro,then we observe and detect morphological changes,apoptosis and expression of apoptosis-related proteins P53 by flow cytometry,confocal laser scanning electron microscopy,enzyme-linked immunosorbent assay(ELISA)and Western blotting(WB),finally use the SPSS 17.0 statistical software for statistical analysis.Results:1,Compared with the healthy group,cell apoptosis rate of hypertension QDBS group,non-QDBS group,different doses of Bu yang huan wu decoction is increased,the difference was statistically significant(P <0.01).In addition,compared with hypertension QDBS group,the apoptosis rate of Bu yang huan wu soup of high,medium and low dose group is lower,the difference was statistically significant(P <0.05).2,After the serum of each group had effected on HUVEC for 24 h,the hypertension QDBS group and non-QDBS group had apoptotic body formation,and healthy group had not.Bu yang huan wu decoction high,medium and low dose groups had different levels phenomenon of nuclear condensation.3,Compared with healthy group,the amount of P53 in hypertension QDBS increased after HUVEC cultured for 24 h,the difference was statistically significant(P <0.01).Compared with hypertension QDBS group,different levels of Bu yang huan wu decoction reduced dose obviously(P <0.05).4,According to the WB test results,the amount of P53 in hypertension QDBS was significantly higher than the healthy group,Bu yang huan wu soup expression of P53 protein in each dose group is less than hypertension QDBS group.Conclusion: The serum of Hypertension QDBS can arouse apoptosis in HUVEC,and serum of Bu yang huan wu decoction effected on hypertension QDBS induce apoptosis.This protective effect may be associated with decrease of apoptosis-related protein P53 expression.
