Effect Of Fuyuan Capsule On Apoptosis And Relevent Gene Expression In Vascular Smooth Muscle Cell Of Rats With Qi-Deficiency And Blood Syasis Syndrome

ObjectiveTo explore the effect of Fuyuan capsule on apoptosis and relevant gene expression in vascular smooth muscle cell of rats with qi-deficiency and blood stasis syndrome and its mechanisms in rat with qi-deficiency and blood syasis .Methods48 SD rats ,18 months old, 40 were choiced accidently. Making rats with qi-deficiency and blood syasis from some methods, such as hunger, swmming, tired , frighten and high fat food,one time every day. After 4 months,they were divided for 5 groups accidently.They are model group , blank control group ,Fuyuan capsule little dose group, Fuyuan capsule middle dose group , Fuyuan capsule large dose group and qi-shen capsule group. Every group has 8 rats. Then give different durg to rats which in Fuyuan capsule little dose group, Fuyuan capsule middle dose group , Fuyuan capsule large dose group and qi-shen capsule group with different dose.The rats in Fuyuan capsule little dose group were gived 0.15g·kg-1 weight Fuyuan capsule. The rats in Fuyuan capsule middle dose group were gived 0.45g·kg^-1 Fuyuan capsule.The rats in Fuyuan capsule large dose group were gived 1.35g·kg^-1 Fuyuan capsule. The rats in qi-shen capsule group were gived 0.52g·kg^-1 qi-shen capsule.The rats in model group and blank control group were gived the same capacity NS.All rats were gived drug one time every day, lasting one month.After giving durg, snip one part of artery. Then observating the configuration of vascular smooth muscle cell with transmission electron microscope and ordinary microscope. The expression of bcl-2,bax protein were determined by immunohistochemical staining.VSMC apoptosis was identified by in situ TdT-mediated dUTP nick end labeling (TUNEL) and smooth muscle cell apoptotic index (AI) was calculated by semi-quantitative analysis.ResultsCompared with blank control group,the model group's expression of Bcl-2 protein was increased(P<0 . 05) , Bax protein was decreased (P<0.05),AI was decreased(P<0.05). Compared with model group,the Fuyuan group's expression of Bcl-2 protein was decreased (P<0.05),Bax protein was increased (P<0.05),the ratio of Bcl-2/Bax were significantly lower,SMA1 was increased significantly(P <0.05).In some extent, Fuyuan capsule middle dose group's result mostly coincides with blank control group. ConclusionRats with qi-deficiency and blood syasis exsists serious disorder of propagation and apoptosis in VSMC. Fuyuan capsule can change the disorder of propagation and apoptosis in VSMC .The mechanism that Fuyuan capsule promote the apoptosis of VSMC in rats with qi-deficiency and blood syasis may be involved the expression balance of Bcl-2/Bax in VSMC.
